Sophora viciifolia Hance,which belongs to Leguminosae,is distributed in most province of china such as Jiangsu,Zhejiang,Hubei,Henan,Shanxi and Gansu.In sophora viciifolia organs,it contains matrine,oxymatrine,oxysophocarpine and other alkali.As a tradition Chinese medicine,it has always been winning widespread attention of people rensently,lots of scholars both from home and abroad are doing more detailed researches on the composition,pharmacologic action and clinical application of matrine and oxymatrine.According to large quantities of documentation,matrine and oxymatrine are two important ingredients that are bioactive in killing insect ,bacteriostasis, anti-inflammatory, immune suppression, anti-heart rate irregularity and anti-tumor.Since 2003,10 medical corporation has been granted to produce new medicine which contains oxymatrine for liverish disease.We need oxymatrine about 50 ton one year. However,national yearly yield still not arrived one ton in currently.So it is very important to exploit new resource instantly.Qinlin and Bashan is one of the most suitable areas for sophora viciifolia growth.Using sophora viciifolia seed as material for exploit oxymatrine is a shortcut.This subject aims at extracting oxymatrine from sophora viciifolia and then make it pure monomer.Frst of all,extracting oxymatrine from sophora viciifolia seeds using ethanol saturate.During the extraction,the yield of matrine is observed to be obviously influenced by solid-to-liquid ratio,time of soak,concentration of ethanol,time of extraction,liquid dosage of deposition,with their effectiveness in order: concentration of ethanol >time of soak >solid-to-liquid ratio> liquid dosage of deposition.Based on experiment concerning one single factor,actor,an orthogonal experiment is carried out to ascertain optimal extraction conditions which prove to be as follows:ethanol (80%),solid-to-liquid ratio (1:4),time of soak (6d) and liquid dosage of deposition.( 6 times).Next,purfied extracting production by silicagel column chromatography,after a process of concentration,elution,and drying,a purity of 95% is obtained. Finaly,take the high-performance liquid chromatography (HPLC) to study the oxymatrine accumulation in the same sophora viciifolia organs and the oxysophocarpine distributing.The results showed that:leaf organ of sophora viciifolia is the main organ for oxymatrine synthesis,seeds is the main organ for oxymatrine accumulation.Early in May the young fruit formed,this is also the turning point oxymatrine transfer from the leaf to the fruits,after the young fruit formed,leaves oxymatrine level in the leaves falling from 1.1 percent to August's 0.5 percent,while oxymatrine level in the young fruit increase from the initial 0.77 percent to mature fruit of 3.6 percent.It also showed the buds were main accumulation organs in ox ysophocarpine,which is highly 2.114%.The contents of oxysophocarpine in other organs such as young fruit,mature fruit,flowers,leaves,one year growing stem and many year-growing stem is respectively1.735%,1.651%,1.615%,0.540%,0.244%.All the result is helpful to the wild resources development and ecology protection of Sophora viciifolia Hance.
