Background Systemic lupus erythematosus (SLE) is an autoimmune disease which complexity of clinical manifestations. Its pathogenesis has not yet been fully clarified which generally believed that genetic, immunity, endocrine and environmental factors, genetic factors play an important role in the pathogenesis of SLE, which related to the development of SLE genetics faster. In the past two decades, many susceptibility genes of SLE had revealed through linkage analysis, candidate gene and the mouse model of lupus. Online Mendelian Inheritance in Man (OMIM) has collected 13 SLE susceptibility loci including 1q41-q42(SLEB1), 2q37.3(SLEB2),4p16-p15.2(SLEB3), 12q24(SLEB4), 13q32(SLEB5), 16q11.2(SLEB6), 20p12(SLEB7), 20q13.1(SLEB8), 1q32(SLEB9), 7q32(SLEB10), 2q32.2-q32.3(SLEB11), 8p23.1(SLEB12) and 6p23(SLEB13), and the last two susceptibility loci using genome-wide association analysis. In recent years, genome-wide association (GWA) study approach as the most effective way to find complex disease susceptibility genes, and a number of SLE susceptibility genes GWA studies had carry out. Harley et al had reported that a single nucleotide polymorphism (SNP) rs1801274 on FCGR2A gene was related to the SLE susceptibility of Europeans (P=6.78×10–7). SLE is a complex disease which is non-Mendelian inheritance patterns with strong ethnic differences and genetic heterogeneity. To investigate the association between FCGR2A gene polymorphism and SLE susceptibility in Chinese Han population. We extracted the data of FCGR2A gene from SLE GWA study results (unpublished), and investigated the association between polymorphism of FCGR2A gene and SLE susceptibility in Chinese Han population. Objective To investigate the association between polymorphism of FCGR2A gene and SLE susceptibility in Chinese Han population. Methods The SNPs were genotyped by the Illumina Human 610-Quad BeadChips in 590 cases of SLE and 1,135 controls, then we selected genotyping data of 5 SNPs which on FCGR2A gene. The association analysis were performed by Plink 1.03 software. Results Association analysis showed that there were no significant differences between the cases and the controls in the allele frequency of 5 SNPs ( rs1801274, rs4657039, rs6696854, rs10800309 and rs511278) on FCGR2A gene (P>0.05).Conclusion The results indicate that there are no association between the 5 SNPs on FCGR2A gene and the susceptibility of SLE in Chinese Han population.
