Studies On The Anti-tumor Effects And Part Mechanisms Of Melittin

Cancer is one of the chief disease that threat people's health .Ahout seven million take these disease per year. about five million people die of cancer .So finding new anti-tumor medcines has been the major research direction in all the world .Melittin that is activity component abstrated from been's venom has strong anti-tumor activity. Most research find that melittin has inhibitory effects to most kinds of tumors in recent years, indicating that melittin has anti-tumor acitivity.But at same time it has some adverse effects including cell toxity justo major. hemolysis .allergy and so on.In this experiment we want to investigate its mechanisms and provide record for the anti-tumor's clincal application of this medicine through observing the changes of cell cycle and immunology index in vivo and in vitro1 screening of melittin in vivo and in vitro1.1 screening of melittin in vitroThe inhibitory effect of melittin on tumor cells proliferation in vitro was evaluated by MTT.Melittin at concertration of 1,2,4,8,16,32,64mg/ml could significantly inhibit the proliferation of human tumor cells in vitro including A549,U2OS,Hep2,HepG2,etal .and these results showed time-effect relationship.and melittin showed more anti-tumor effect to human liver cancer cells than other cells. 1.2 screening of melittin in vivoMelittin diaplay significantly inhibitive effects to H22 tumor- burdened mice in the experiment of H22 tumor-burdened mice,while the effects enhanced along with the increase of melittin's concentration indicating that maybe melittin has strong anti-tumor activity.2. Studies on the anti-tumor mechanisms of melittin2.1 Effects of melittin on the cell cycle of H22 cellThrough observing the changes of cell cycle after melittin was intributed to H22 cell . we find that melittin could blot cell S cycle.and apoptosis peak could be observed through flow cytometer. At the same time we could find that H22 cell show the apoptosis characteristic through Hochest dyeing2.2 Effects of melittin on immunue function with tumor-bearing miceThe results in vivo indicate that melittin could increase the lymphocyte proliferation that induced by ConA,inhance the immunue level at doses 2 and 4μg/ml.at the same time increase the anti-tumor rate and suvival time of tumor- burdened mice, raise the Spleen index and IL-2 , CD4+,CD8+ Cell content but reduce TNF-αproduction .And all these finding showed dose- independent relationship.suggesting that the effects of melittin might concern with the strengthen of tumor- burdened mice...
