Study On The Absorption And Metabolism Of Raddeanin A

Rhizoma Anemones Raddeanae, the rhizome of Anemone raddeanaRegel(Ranunculaceae), is commonly used in China folk medicine for curingrheunmatism and neuralgia.Raddeanin A ,a triterpenoid saponin isolated from Anemone raddeana Regel,has good antitumor activity without side effect of increasing the frequencyof SCE in vitro as well as in vivo by ip and ig. In this thesis, the absorptionand metabolism of raddeanin A in vitro and vivo are reported.When injected with Raddeanin A, the metabolites such asβ-D-glucopyranosyl-(1→2)-α-L-arabinopyranosyl-oleanolic acid, oleanolic acidcan be found in the blood of the rats. When intragastric administration withRaddeanin A, besides the metabolites motioned above, an unknown metaboliteRX can be tested.After intragastric administration with Raddeanin A, the urine was collected.Raddeanin A,β-D-glucopyranosyl-(1→2)-α-L-arabinopyranosyl-oleanolic acidand oleanolic acid can be found in the urine.We also studied the main metabolism of raddeanin A in liver homogenate of ratsby TLC and MS. In homogenized liver, the main metabolites of raddeanin A areobtained by hydrolysis, and in homogenized liver induced by Phenobarbital, themain metabolite may be obtained by reduction and fixation reaction.The absorption of Raddeanin A was studied with the model of everted sac of rat'sjejunum. The result showed that it was hard to be absorped.
