Objective: To investigate the effect of allitride injection preconditioning and ischemic preconditioning on renal ischemia reperfusion injury in rats.Methods: To establish rat's I/R injury model. A total of 75 rats were randomized into five groups (n=15, in each). Control group(CON), I/R control group(IR), ischemic preconditioning group(IPC), allitride preconditioning group(APC), allitride preconditioning and ischemic preconditioning group (AIPC). Operation approach: Resecting the right kidney without occluding the left renal artery. IR group:Ischemia was induced by applying the left renal artery occluded for 45 minutes followed by reperfusion on the basis of the right renal resection. IPC group: Resecting the right kidney , occluded the left renal artery for 8 minutes then reperfusion for 5 minutes at circulate four times, after this ischemia was induced by applying the left renal artery occluded for 45 minutes followed by reperfusion; APC group: 8 H,4 H before operation and each 8 H after operation allitride 20mg/kg were administered via abdominal cavity injection for a total of five doses, whose method of operation is identical with the IR group; AIPC group: 8 H,4 H before operation and each 8 H after operation allitride 20mg/kg were administered via abdominal cavity injection for a total of five doses, whose method of operation is identical with the IPC group ; Kidney tissue and blood specimen Were collected by all groups 24 hours after operation. Changes of the renal pathological morphology was observed by hematoxylin-eosine(HE) staining, Levels of serum creatinine(Cr), urea nitrogen(BUN) were determined by automatic biochemistry analyzer; Levels of malonaldehyde(MDA) were determined by Thibabituric Acid(TBA); Levels of superoxide dismutase(SOD) were determined by xanthine oxidase(XO);Intercellular adhesion molecule-1 (ICAM-1) expressions in kidney were tested by immunohistoe-hemistry.Results: 1.Renal tissue pathological change: Control group: Not detected obviously abnormality; IR group: The injury in renal tissue is manifest; IPCgroup, APC group and AIPC group: The injury in renal tissue is lighter than IR group.2.Renal function:.Compared with CON group, the level of BUN and Cr in IR group that remained higher value(P<0.01) , and it was distinctly lower in IPC group and APC group and AIPC group than that in IR group (P<0.01); Compared with AIPC group, the level of BUN and Cr in IPC group and APC group that remained higher value(P<0.01); Compared with IPC group, the level of BUN and Cr in APC group that was not distinctly difference(Pï¹¥0.05).3.In CON group , level of blood serum MDA to maintain in low level; Compared with CON group, the level of MDA in IR group that remained higher value(P<0.01), and it was distinctly lower in IPC group and APC group and AIPC group than that in IR group (P<0.01); Compared with AIPC group, the level of MDA in IPC group and APC group that remained higher value(P<0.01); Compared with IPC group, the level of MDA in APC group that was not distinctly difference(Pï¹¥0.05).4.The level of blood serum SOD in CON group to maintain in normal level; Compared with CON group, the level of SOD in IR group that was cut manifest down (P<0.01), and it was distinctly higher in IPC group and APC group and AIPC group than that in IR group (P<0.01); Compared with AIPC group, the level of SOD in IPC group and APC group that remained lower value(P<0.01); Compared with IPC group, the level of SOD in APC group that was not distinctly difference(Pï¹¥0.05).5.The level of renal tissue ICAM-1: There has no obviously ICAM-1 express in light microscope of CON group; In IR group, ponderosus ICAM-1 express in renal tissue, especially in nephric tubule endothelial cell,other place has little espress; The ICAM-1 espress is low in IPC group and APC group, In AIPC group , the ICAM-1 espress is lower than IPC group and APC group. Mean Optical Density(MOD): The MOD value in IPC group and APC group is higher than CON group(P <0.01) and AIPC group(P <0.01),but lower than IR group(P <0.01);. Compared with IPC group, the MOD value in APC group that was not distinctly difference(Pï¹¥0.05).Conclusion: 1. Pathological animals subjected to renal ischemia showed characteristic disruption of architecture and renal dysfunction.Ischemia for 45min followed by reperfusion leads to renal ischemical-reperfusion injury, however the damage is reverse.2.The level of blood serum MDA obviously step up after kidney ischemia reperfusion injury, there are positive correlation between MDA and renal function . to illustration there are relate between MDA and kidney ischemia reperfusion injury.3.The level of blood serum SOD obviously breakdown after kidney ischemia reperfusion injury, there are positive correlation between SOD and renal function . to illustration there are relate between SOD and kidney ischemia reperfusion injury.4.The level of ICAM-1 express in kidney tissue obviously step up after kidney ischemia reperfusion injury, there are positive correlation between ICAM-1 and renal function. to illustration there are relate between ICAM-1 and kidney ischemia reperfusion injury.5.Ischemic preconditioning or allitride preconditioning would protect kidney ischemia reperfusion injury,Probably it proctects kidney through lower MDA level and enhance SOD activity.6 . Ischemic preconditioning or allitride preconditioning would protect kidney ischemia reperfusion injury,and that the two factor would have synergy in protect kidney ischemia injury reperfusion. Probably it pro- tects kidney through lower ICAM-1 expressions.
