Objective To study the effects of dipyridamole preconditioning on the expression of intercellular adhesion molecule-1(ICAM-1)in the ischemia-reperfusion injury of liver sinusoidal endothelial cells(LSEC)in rats.Methods Using ischemia- reperfusion rat model in normal temperature.Thirty-six rats were randomly divided into three groups(12 rats in each);group S:sham operation group;group I/R: ischemic-reperfusion model;group P:pretreated the ischemic-reperfusion model with dipyridamole.Ischemia and reperfusion were produced by clamping the hepatic artery and portal vein with a clamp for 60min,then reperfusing for 0.5 hours,1 hours and 2 hours.The liver tissues and blood samples were collected after hepatic portal clamping for 30 min(T1),0.5 hours after reperfusion(T2),1 hours after reperfusion(T3),and 2 hours after reperfusion(T4) respectively.The changes of MDA,ALT,AST were observed.The expression of ICAM-1 protein was measured by immunohistochemical.Examine the ultrastructure of liver with electron microscope.Results①Compared to group S,in groups I/R and P,the injury of liver was found by pathological examination:the levels of serum ALT,AST and ICAM-1 expression of liver tissues were significantly higher(P<0,05);The serum ALT and AST levels and ICAM-1 expression in group P were much lower than those in group I/R(P<0.05).②The MDA content of liver tissues in group P were much lower than those in group I/R in T2~T4(P<0.05).③Electron microscopic examination showed that the sinusoidal endothelial cell seriously damaged in group I/R and dipyridamole significantly ameliorated the injury of liver induced by ischemia-reperfusion. Conclusion Preconditioning with dipyridamole can protect liver sinusoidal endothelium from I/R injury in rats.
