Objective:To investigate the anti-fungus mechanism of Potassium iodide against S.schenckii and the difference of anti-S. schenckii ability cooperate with different immunocytes.Methods:The experiment divided into eight groups which composed with KI, spore suspension and different immunocytes,those groups are Lym+KI+SS(I),MNC+KI+SS(Ⅱ),Lym+MNC+KI+SS(Ⅲ),PMNs+SS(Ⅳ),PMNs+KI+SS(Ⅴ),PMNs+MNC+KI+SS(Ⅵ) ,PMNs+Lym+KI +SS(VII),PMNs+Lym+MNC+KI+SS(Ⅷ),after incubate two hours, take same quantity mix-solution from each group and inoculate the solution on SDA, count the numbers of clonal formation unit to explore the difference of anti-S. schenckii ability, conclude which immunocyte exert the effect of anti-S. schenckii。For the sake of define the mechanism more clear, we detect the production of reactive oxygen species of the polymorphanuclear neutrophils in the groups which with-Leukomonocyte and no-Leukomonocyte,and the polymorphanuclear neutrophils that labeled with DCFH-DA,the level of reactive oxygen species measured by laser confocal microscopy to discuss the mechanism. Results:In the groups of single immunocyte which only contain PMN(sⅤandⅥ)or Lym(Ⅰ)or MNC(Ⅱ),the number of CFU respectively is 383±26,379±23,766±36,748±27,the group ofⅤandⅥhave significant difference to the group ofⅠorⅡ(P<0.01),but there is no prominent difference between the groups which contain KI(Ⅴ) or no KI(Ⅳ) (P>0.05).There is also no significant difference between the groups of Lym and MNC(P>0.05). In the groups of multi-immunocytes, the numbers of CFU in the groups(Ⅵ,Ⅶ,Ⅷ) which contain PMNs is 379±23,1 24±21,1 27±20,which less than no PMNs groups (Ⅲ: 739±41 ) ( P<0.01 ) , and the CFU in the groups (Ⅶ:124±21,Ⅷ:127±20)which contain PMNs and Lym simultaneously are less than no Lym group(Ⅵ:379±23), the difference has prominent significance(P<0.01).The number of CFU in the groups which contain MNC (Ⅵ:379±23 andⅧ127±20) has no significant difference to the groups which no MNC(Ⅴ:383±26 andⅦ:124±21). In the next experiment, we found the level of ROS in the PMNs which activated by SS wound increase during 12 hours, and wound fall obviously at 24 hour. No matter have Lym or not,there is no obvious difference of the level of ROS in PMNs between the two groups in different times(P>0.05).Conclusion : KI can activate Leukomonocyte to secrete some cytokine and it can promote the anti-S. schenckii ability of polymorphanuclear neutrophils,but not by regulate the level of reactive oxygen species in polymorphanuclear neutrophils to implement the effect.
