OBJECTIVETo investigate the expression law of endogenous zinc finger protein A20 of mouse lung tissues in the process of spinal cord injury (SCI)and(or) lipopolysaccharide(LPS) infection.METHODSA total of 220 healthy mice(from Kunming,Yunnan province) were randomized into four groups:control group,SCI group,LPS group and SCI plus LPS group.In lung tissues, the expression of A20 protein and mRNA was measured by immunohistochemistry(IHC) and reverse transcription-polymerase chain reaction (RT-PCR) ,and the degree of pathological lesion was tested by HE staining.RESULTS1. In group control , A20 protein expressed at a low leve1(0.142±0.012). In group SCI, the expression of A20 protein was significantly higher than group control during 4~48 hours(P<0.05).In group LPS, high A20 protein expression began to appear 0.5 hour(0.202±0.020), significantly higher than group control during 0.5~24 hours (P<0.05)and group SCI during 0.5~8 hours(P<0.05). The highest expression of A20 protein was tested in group SCI plus LPS, significantly higher than group control during 0.5~48 hours(P<0.05),group SCI during 0.5~8 hours (P<0.05)and group LPS during 8~48 hours(P<0.05).2. In group control , the A20 mRNA expressed at a low leve1(0.883±0.033). In group SCI, the expression of A20 mRNA was significantly higher than group control during 4~24 hours(P<0.05).In group LPS, the expression of A20 mRNA significantly higher than group control during 0.5~24 hours (P<0.05)and group SCI during 0.5~4 hours(P<0.05). The highest expression of A20 mRNA was tested in group SCI plus LPS, significantly higher than group control during 0.5~48 hours(P<0.05),group SCI during 0.5~24 hours(P<0.05) and group LPS during4~48 hours(P<0.05).3. In group SCI plus LPS,severe pathologic damage of lung tissues was observed during 0.5~4 hours. The lung tissues emerged neutrophil infusion ,epithelial cell swelling and so on. the pathologic damage was alleviated after 8 hours ,but still severer than it in group control .CONCLUSIONSIn the early period of SCI,zinc finger protein A20 expresses high especially with endotoxemia.It is indicated that increased expression of A20 in the tissues following SCI with endotoxemia may play an important role in regulating inflammatory response.
