The Study Of Doxycycline United Fluorouracil Inhibiting Liver Metastasis In Colorectal Carcinoma

Objective:To investigate the effects of matrix metalloproteinases(doxycycline) united fluorouracil(5-FU) on the inhibition of colorectal cancer liver metastases.Methods:To set up liver metastasis model of human LoVo colorectal cancer:human LoVo colorectal cancer cells are cultured in the modified RPMI-1640 culture medium containing 10%FBS,digested,after centrifugal adjusted the concentration of cells to 1×10~6/ml and injected to 40 nude mice spleens.The mouses are randomly divided into four groups:saline group,5-Fu group,doxycycline group,5-Fu united doxycycline group.After feeded 3 weeks,to be observed the number of metastatic carcinoma of the liver and spleen of tumor size.Results:1.The size of spleen tumor:spleen tumor size of saline group,5-Fu group,doxycycline group,5-Fu united doxycycline group are 0.83±0.21cm~3,0.48±0.12cm~3,0.71±0.18cm~3,0.27±0.10cm~3.spleen tumor size of 5-Fu group,5-Fu united doxycycline group are smaller than the saline control group(P＜0.05);there is a significant difference in tumor size of 5-Fu group and the doxycycline group(P＜0.05);spleen tumor size of 5-Fu united doxycycline group are smaller than 5-Fu group(P＜0.05);there is no significant difference in spleen tumor size of doxycycline group and the saline control group(P＞0.05).2.The number of hepatic metastases:the respectively number of Saline groupⅠ,Ⅱ,Ⅲ,class is 1,2,7.the respectively number of 5-Fu group and doxycycline groupⅠ,Ⅱ,Ⅲ,class is 3,6,1.the respectively number of 5-Fu united doxycycline groupⅠ,Ⅱ,Ⅲ,class is 9,1,0.the number of metastatic tumors of 5-Fu group,doxycycline group,5-Fu united doxycycline group are less than saline control group(P＜0.05);there is no significant difference in the number of metastatic tumors of 5-Fu group comparing to the doxycycline group(P＞0.05);the number of metastatic tumors of 5-Fu united doxycycline group comparing to 5-Fu group and doxycycline group are less(P＜0.05).Conclusions:1,Doxycycline has a weak role in the inhibition of the spleen tumor, however,doxycycline can be markedly enhanced fluorouracil on the inhibition of tumor cells growth.2,Doxycycline can inhibit colorectal cancer cell invasion and metastasis,moreover can be markedly enhanced fluorouracil on the inhibition of tumor cells invasion and metastasis.
