ObjectivesIn this study , the selective inhibition of artemisinin combined with ferrous succinate on the human hepatoma SMMC-7721 cells was investigated and the expression of transferrin and its receptor in the cancer tissues of the control group was detected.Methods1. The establishment of animal model. The animal model of mouse-borne tumor was made by subcutaneous transplanting human hepatoma carcinoma SMMC-7721 cells to the waist of mouse.2. Experimental grouping and intervention. The tumor bearing mice were randomly divided into 4 groups and intervened with artemisinin, ferrous succinate, artemisinin plus ferrous succinate, physiological saline (control group), respectively, and gross tumor volume was observed.3. Tumor inhibition rate was evaluated. Record the maximum tumor diameter, diameter and then gross tumor volume was calculated; check the quality of tumor to calculate the tumor inhibition rate.4. Apoptosis of tumor cell was detected by TUNEL . Apoptosis of tumor cell was detected by in situ end-termination method (TUNEL) on the paraffin sections of tissue, the apoptosis index (AI) was calculated.5. The expression of transferrin and its receptor on the cancer tissues was detected by SP immunohistochemical assay. The expression of transferrin and its receptor on the paraffin section of cancer tissues was detected respectively by immunohistochemical SP method ;the results was observed and recorded under the microscope.Results1. The animal model of mouse-borne tumor can be made by subcutaneous transplantation,and its success rate was 72.5%. 2. Compared with physiological saline, artemisinin, artemisinin plus ferrous succinate obviously suppressed tumor growth of tumor-bearing mice(P<0.05), the tumor inhibition rate of which was 33.21%, 55.04% respectively, which showed significant difference(P<0.05).3. Compared with physiological saline, artemisinin, artemisinin plus ferrous succinate induced apoptosis of tumor cells, and apoptotic indexs were 13.13% and 31.35%, which had statistical significance compared with control group(P<0.05).4. Transferrin and its receptor on the hepatocellular carcinoma issues in the control group and ferrous succinate group was high expression, which had statistical significance compared with artemisinin, artemisinin plus ferrous succinate groups(P<0.05). But compared with the before and after two groups, it had no statistically significant differences(P>0.05).ConclusionsThe subcutaneous transplantational model can be a successful model,and it may meet the basic requirements of this experiment.Both artemisinin and artemisinin combined with ferrous succinate had the effct on inhibition growth and promotion apoptosis of tumor cells, but the role of a stronger combination.
