Objective : To investigate the effect of down-regulation CD133 expression by RNA interference on the cell proliferation and invasion of human hepatoma carcinoma SMMC-7721 cells.Methods:The recombinant eukaryotic expression vectors targeting CD133 were constructed in vitro and transfected into SMMC-7721 cells and RT-PCR and Western blot were used to detect the expression of CD133 in SMMC-7721 cells. The cell apoptosis was evaluated by flow cytometry and the proliferation and invasion of SMMC-7721 cells were determined by MTT assay and Transwell assay respectively..Results : The recombinant eukaryotic expression vectors PGPH1/GFP/Neo-shGAPDH, PGPH1/GFP/Neo-shNC, PGPH-1/GFP/Neo-CD133-1 and PGPH1/GFP/Neo-CD133-2 were successfully constructed in vitro and transfected into SMMC-7721 cells. Compared with untransfected and nonspecific(PGPH1/GFP/Neo-shNC) transfected SMMC-7721 cells, the expression of CD133 in PGPH1/GFP/Neo-CD133-1 and PGPH1/GFP/Neo-CD133-2 transfected SMMC-7721 cells were inhibited significantly at both mRNA and protein levels, at an inhibitory rate of 32.98% and 76.16% for CD133 mRNA and 49.46% and 59.15% for CD133 protein. Compared with untransfected and nonspecific transfected SMMC-7721 cells, the cell apoptotic rate was increased significantly in SMMC-7721 cells trasfected with PGPH1/GFP/Neo-CD133-2; the proliferation and invasion of SMMC-7721 cells transfected with PGPH1/GFP/Neo-CD133-2 were both inhibited markedly.(P<0.05)Conclusion : RNA interference targeting CD133 down-regulates expression of CD133, induces cell apoptosis, inhibits cell proliferation and invasion in human hepatoma carcinoma SMMC-7721 cells.
