Objective:1,To investigate the expression of BNIP3 protein in colorectal cancer tissues,adenoma of colon,and normal colorectal mucosa tissueTo investigate their role in the occurrence and development of colorectal cancer.2,To investigate the correlation of BNIP3 protein with the prognosis of colorectal cancer,and to evaluate their value for prognosis of the patients with colorectal cancer.Methods:1,To collect 70 primary colorectal cancer cases and 20 colorectal adenoma cases during July 1,2001 and July 31,2003 in cancer hospital of Hunan Province,which were all treatmented by operation and confirmed by pathology results and their paraffin-embedded specimens were saved very well.All the cancer cases were not accept chemotherapy or radiotherapyImmunohistochemical technique was used to detected the expression of BNIP3 protein in the paraffin-embedded specimens of 70 colorectal cancer cases,normal Colorectal mucosa Corresponding with the cancer and 20 colorectal adenoma cases.2,The software package spss 13.0 was used for statistical analysis.The expression of BNIP3 protein in different groups was analysed by X~2 test,Kaplan-Meier and Log-rank test were used to calculate and analyse survival rate.Prognostic factors was analysed by COX model.Result:1,The BNIP3 protein mainly expressed in cytoplasm of colo rectal cancer cell,partly expressed in nucleus.The positive expression rate of BNIP3 protein was 57.14%in 70 colorectal cancer cases and 40.00% in normal colorectal mucosal tissues,while 85.00%was in 20 colorectal a denoma tissues.There was a significant difference between the three groups (p<0.05).2,By statistical analysis,the expression of BNIP3 was found to be associated with different types of histology of colorectal cancer: The positive expression rate of poorly differentiated adenocarcinoma(18. 75%)was significantly lower than moderately-differentiated adenocarcinoma (66.67%)and well-differentiated adenocarcinoma(83.33)(p<0.05).But the well-differentiated adenocarcinoma had no difference with moderately differentiated-adenocarcinoma(p>0.05).3,By statistical analysis,the expression of BNIP3 protein has no correlation with the clinicopatholog ic factors such as age,sex,location,Infiltration,lymph node metastasis, (p>0.05);but has correlation with Dukes stage,differentiation of aden ocarcinoma(p<0.05) 4,The total five-year survival rate was 76.00%in 70 cases of patients.The five-year survival rate after surgery in the patients with positive expression rate of BNIP3(87.50%)was significantly higher than the negative one(63.30%).which had a significant difference (p<0.05).Multivariate analysis showed that BNIP3 was an protective factor. Its Odds ratio(OR) was 0.743,But wasnot an independent prognostic factor for patients with colorectal cancer(p>0.05).Conclusion:1,The positive expression of BNIP3 protein in colorectal cancer tissues was significantly higher than that in normal colorectal mucosal tissues.but significantly lower than its expression in colorectal adenoma tissues.2,The expression of BNIP3 protein in colorectal cancer tissues associated with different types of histology and Dukes stage;but no correlation with the clinicopathologic factors such as age,sex,location, Infiltration,lymph node metastasis.3,BNIP3 was an protective factor, But was not an independent prognostic factor for patients with colorectal cancer,can not be used as an independent prognostic factor for patients with colorectal cancer.
