Objective To investigate the killing effect of arsenic trioxide (AS2O3) in vitro on brain tumor stem cells for further animal experiments and clinical trials to provide the theoretical basis.Expect to find a new,feasible and effective treatment.Methods To culture brain tumor stem cells with U251 glioma cells.To compare the sensitivity of U251 glioma cells and brain tumor stem cells to AS2O3,ACNU with MTT test and Flow cytometry.To observe of cell morphological changes with haematoxylin and eosin,Hochest staining.Results We induced brain tumor stem cells by the use of U251 glioma cells.MTT test and Flow cytomety showed that U251glioma cells were both sensitive to AS2O3 and ACNU.They can both inhibit the growth of tumor cells obviously.The biggest U251 apoptosis rates were 58.37%,72.85%in MTT test.ACNU is greater compared to AS2O3.AS2O3,ACNU were both drug resistence on brain tumor stem cells,they can only kill a small number of brain tumor stem cells.The biggest brain tumor stem cells apoptosis rates were 21.81%,21.72%in MTT test,No significant difference between the two compared.Conclusion We can induce brain tumor stem cells by the use of U251glioma cells.AS2O3 can inhibit the growth of U251 glioma cells obviously,but ACNU is greater compared to AS2O3;AS2O3,ACNU were both drug resistence on brain tumor stem cells obviously.No significant difference between the two compared.Compared to tumor cells,brain cancer stem cells more resistant.
