OBJECTIVE:To isolate,culture and identificate of brain cancer stem cells from glioma cell line U87.METHODS:To culture the brain tumor cell line U87 in serum-free medium with EGF, bFGF,N2 and B27,the immunofluorescence staining was employed to identificate the brain cancer stem cells and the differentiated cells.RESULTS:The brain cancer stem cells from glioma cell line U87 have been successfully isolated.They formed typical neurospheres in serum-free medium,and had strong capacity to self-renew,and proliferate.The cells expressed CD133 and Netin,They could differentiate into cells that expressed GFAP.CONCLUSION:The brain cancer stem cells exist in the glioma cell line U87,and they can be isolated,cultured and differentiated.
