Studies Of The Relation Between STAT3 And Invasion And Metastasis Of Choriocarcinoma

Objective:To explore mechanism and relationship between the expression of signal transducers and activators of transcription 3 (stat3) and the invasiveness, metastasis of choriocarcinoma cells.Method:(1)Choriocarcinoma cell lines(JEG-3 cells and JAR cell )was cultured in vitro and the human chorionic villi tissue was collected(2) The invasiveness was assessed by penetration through a matrigel-coated filter in transwell chamber ability between JEG-3 cells and JAR cell(3) Expression of stat3 and p-stat3 protein in the human chorionic villi tissues and choriocarcinoma cell lines(JEG-3 cells and JAR cells) were detected by immunocytochemistry and western blot.(4) The correlation of stat3 and p-stat3 protein expression level and the level of choriocarcinoma cell invasion in vitro was analyzed by correlation analysis of binary variablesResult: (1)The expression of stat3 and p-stat3 protein can be found in choriocarcinoma cell lines (2) The mount of JEG-3 cells penetrating through matrigel-coated filter in transwell chamber was significantly increased compared with JAR cells under light microscope (P<0.05)(3) SP immunocytochemical result showed the expression of stat3,p-stat3 significantly increased in JEG-3 cells compared with JAR cells; Protein expression level of stat3,p-stat3 also signifieantly increased in JEG-3 cells compared with JAR cells by western blot, and markedly stronger in choriocarcinoma cell lines compared with normal villi tissues(p<0.05,p<0.01) (4) Correlation analysis of binary variables show stat3 protein expression level and the level of choriocarcinoma cell invasion in vitro was positively correlated (r=0.81, r=0.78, p<0.01) .Conclusions: (1)The expression of stat3 and p-stat3 protein was higher in choriocarcinoma cells than normal villi tissues,especially higher in JEG-3 cells compared with JAR cells,so it showed that stat3 led corresponding increase of p-stat3 and probabilly contributed to the invasion and metastasis of choriocarcinoma cells.(2)There were significantly positive correlations between stat3,p-stat3 and the invasiveness of choriocarcinoma cells,so it showed that stat3 and p-stat3 were relative to the development of choriocarcinoma.
