[Objective]To investigate relationship between the expression of signal transducer and activator of transcription3 (STAT 3), phosphorylation Signal transducer and activator of transcription3(P-stat3) Hypoxia-inducible factor-1α(HIF-1α), Vascular Endothelial Growth Factor(VEGF) and invasiveness and metastasis in non-small cell lung cancer.[Methods]Immunohistochemistry were used to detect the expression of STAT 3-, p-Stat3N HIF-1αand VEGF in non-small cell lung cancer and their normal counterparts. The association between STAT 3,p-Stat3,HIF-1α,VEGF expression and clinicopathologic parameters was analyzed.[Results]1. stat3 p-stat3,HIF-1 a and VEGF were higher expression in lung cancer tissues than in lung peficancerous tissues,χ2 and p respectively(2=5.431, P=0.02;,χ2 =10.820, P<0.001,,χ2=12.000, P<0.001;χ2=19.513, P<0.001), There were statistical differences about the four protein expression between lung cancer tissues and lung peficancerous tissues.2. there exists a positive relation between every two among stat3 p-stat3,HIF-1aand VEGF (Kruskal-Wallis H), P value were less than 0.01, r and p respectively(Stat3 and p-stat3, r=0.468,P=0.000; Stat3 and VEGF, r=0.430, P=0.000; Stat3 and HIF-1a, r=0.329, P=0.007; p-stat3 and VEGF, r=0.732, P=0.000; p-stat3 andHIF-1a, r=0.616, P=0.000; VEGF and HIF-1a, r=0.802, P=0.000).3. Overexpression of stat3,p-Stat3,HIF-la and VEGF were significantly associated with the presence of metastatic lymph nodes,χ2 and p respectively(χ2 =9.405 P=0.002;χ2=11.259 P=0.001;χ2=10.864 P=0.001;χ2=19.175 P =0.000), P value were less than 0.01, There were statistical differences about the four protein expression between lung cancer tissues and lung peficancerous tissues.furthermore, stat3,p-Stat3,HIF-1a and VEGF protein expression have a close relation with clinical stage,χ2 and p respectively(χ2=15.058, P=0.002;χ2=16.719,P=0.001;χ2=14.763, P=0.002;χ2=27.763<0.001),and have a close relation with differentiation; r and P respectively (r=-0.422, p=0.000; r=-0.416, P =0.001; r=-0.382, P=0.002; r=-0.359, P=0.003), P value were less than 0.01.4. Stat3,p-Stat3,HIF-1a,VEGF protein expression in lung cancer with the histologic typeχ2 and p respectively (χ2=13.790, p=0.087;χ2=14.987, p=0.060;χ2=7.794, p=0.453;χ2=13.786, p=0.088),sexχ2 and p respectively (χ2=3.594, p=0.425;χ2=5.551, p=0.215;χ2=3.182, p=0.532;χ2=4.197, p=0.339),and ageχ2 and p respectively (χ2=8.247, p=0.083;χ2=5.547, p=0.219;χ2=7.878, p=0.082;χ2=7.399, p=0.092). p value all above 0.05. Stat3,p-Stat3,HIF-1a,VEGF protein expression have no statistically significant difference.[Conclusion]1. Compare with negative expression in normal counterparts, a higher Stat3,p-Stat3,HIF-1α,VEGF expression was detected in non-small cell lung cancer and associated with invasiveness and lymph node metastasis, and Stat3,p-Stat3,HIF-1α,VEGF are considered working as factors indicating a poor prognosis clinically in non-small cell lung cancer. 2. There exists a positive correlation about stat3,p-stat3,HIF-1α,VEGF protein expression with multiple comparison, show up a positive regulation among stat3. p-stat3,HIF-1a,VEGF protein expression. Stat3^ p-Stat3,HIF-1 a in conjuncting with VEGF, promote invasiveness and metastasis of non-small cell lung cancer.It can be supposed that block expression of any of these four factor especial VEGF can suppress invasiveness and metastasis of non-small cell lung cancer.
