Objective:To investigate membrane-cytoskeleton linker Ezrin abnormal activation in the Human Epidermal Carcinoma Cell Line,and identify the effect of Ezrin on the growth and motility of Human Epidermal Carcinoma Cell Line A431.Investigate the effect of Baicalein on the growth of Human Epidermal Carcinoma Cell Line through the Ezrin.Methods:1,The expression of Ezrin and Phos-Ezrin(phosphorylated-Ezrin) was detected in 36 cases of squamous cell carcinoma(SCC),27cases of basal cell carcinoma(BCC),20 cases of seborrheie keratosis(SK) and 10 cases of normal skin(NS) using immunohistochemistry.2,Non-cytotoxic concentration(NCC) of Baicalein was measured on A431 using MTT assay.After treatment with Baicalein at NCC,the expression of Ezrin and phos-Ezrin was detected in A431 cell using Western-blotting,and pseudopod formation of A431 cell was observed using scanning electron microscope.The inhibitory effect of Baicalein on A431 motility was determined using wound healing assay.Results: 1,The positive rates of Ezrin and Phos-Ezrin in NS,SK,BCC, SCC were 20.0%and 10.0%,25.0%and 15.0%,66.7%and 77.7%, 91.3%and 97.2%,respectively.There was significant difference between BCC and NS(p<0.01),SCC and NS(p<0.01).2,Ezrin and phos-Ezrin expression increased alongwith the clinic grade of SCC.The positive rate of Ezrin and phos-Ezrin in SCC with lymphatic metastases was higher than that one with non-metastses (P<0.01).3,MTT assay data showed that Baicalein could inhibit the proliferation of A431 at the concentrations greater than 0.5μg/ml(P<0.05), and no effect at less than 0.5μg/ml.The NCC of Baicalein on A431 was 0.1~0.5μg/ml.Western blotting results showed that Baicalein could inhibit phos-Ezrin expression at dosage-dependent,eseudopod of A431 cells obviously decurtated and its amount reduced after Baicalein treatment.Baicalein also suppressed a migration of A431 cells.Conclusion:1,The highly-expression of Ezrin and Phos-Ezrin is closely associated with skin tumor clinic grades and lymph node metastasis,and maybe is an important molecular event to the pathogenesis of SCC.2,Baicalein can inhibit Ezrin phosphorylation at a non-cytotoxic concentration,not total Ezrin.3,Baicalein may inhibit the invasion,motility and pseudopod formation of A431 cell through inhibiting Phos-Ezrin,and finally inhibits the metastasis of epidermal carcinoma.
