[Objective] To investigate the expression of Ezrin in protein and gene level in cervical cancer and their relationship with clinical feature,especially with the invasion and metastasis of cervical cancer surgery patients.In order to find a valuable marker for condition and prognosis monitoring in cervical carcinoma and provide theoretical basis of therapeutic target at the same time.[Methods] Collecting 15 cases of normal cervical tissues, cervical intraepithelial neoplasia tissues and invasive cervical carcinoma stageⅠto stageⅢrespectively. Using reverse transcriptase polymerase chain reaction (RT-PCR) method detect the Ezrin mRNA expression of each group tissues,and using immunohistochemical SP method detect the Ezrin protein expression.Statistics was done by SPSS 17.0 in which Chi-square test or correcting chi-square was used for rate data, nonparametric statistics Kruskal-Wallis rank sum test or Mann-Whitney U test was used for ranked data.The correlation between the Ezrin mRNA and protein in cervical carcinoma was tested by pearson analysis, significance was considered if P<0.05.[Results]1. RT-PCR method showed:The levels of Ezrin mRNA in invasive cervical carcinoma stageⅠ,Ⅱ,Ⅲwere higher than the levels of normal cervix, cervical intraepithelial neoplasia. The expression of each group with median was respectively:normal cervical 0.09, cervical intraepithelial neoplasia 0.19, cervical carcinoma stageⅠ1.55, cervical carcinoma stageⅡ1.13, cervical carcinoma stageⅢ 1.65.Comparing five groups through non-parameter statistics Kruskal-Wallis rank, the differences were statistically significant (Z=19.938, P=0.001), it means that Ezrin mRNA expression levels were not all the same among five groups (P<0.05). Pairwise comparison results showed that levels of Ezrin mRNA expression in invasive cervical carcinoma stageⅠ,Ⅱ,Ⅲwere significantly higher than the group of normal cervix and cervical intraepithelial neoplasia,the difference was statistically significant (P<0.05). Other groups were not statistically significant (P>0.05). Ezrin mRNA expression levels in different pathological grade,pathological type, tumor size,age were not statistically significant (P>0.05),but it has significant difference in the depth of cervical stromal invasion and lymphnode metastasis in cervical carcinoma surgery patients (P<0.05).2. Immunohistochemistry showed:The expression levels of Ezrin protein in normal cervix, cervical intraepithelial neoplasia and invasive cervical cancer stageⅠ,Ⅱ,Ⅲwere different, the positive expression rates respectively were 26.68%(4/15),46.68% (7/15),60.00%(9/15),80.00%(12/15) and 93.33%(14/15). Through chi-square test, Ezrin protein expressions among five groups were not the same (χ2=17.654, P< 0.05). Pairwise comparison results showed that the levels of Ezrin protein expression in invasive cervical carcinoma stageⅡ,Ⅲwere significantly higher than the group of normal cervix,the differences were statistically significant (P<0.05), expressions between the group of cervical intraepithelial neoplasia and cervical carcinoma stageⅢwere statistically different (P<0.05). The differences of other groups were not statistically significant (P>0.05). Ezrin protein expression levels in the depth of cervical stromal invasion and lymphnode metastasis were statistically significant in cervical carcinoma surgery patients (P<0.05),but no significant difference in different pathological grade,pathological type, tumor size,age in invasive cervical carcinoma (P>0.05).3. Correlated analysis between Ezrin mRNA and Ezrin protein results showed:Ezrin mRNA and Ezrin protein expression in cervical cancer tissues were related (r=0.508. P=0.001). Ezrin protein and Ezrin mRNA expression were positive correlation. [Conclusions]1. Ezrin mRNA and Ezrin protein expression can be detected in cervical cancer tissues, cervical intraepithelial neoplasia tissues and normal cervical tissues, The levels of Ezrin mRNA expression in invasive cervical carcinoma stageⅠ,Ⅱ,Ⅲwere higher than the levels of normal cervix, cervical intraepithelial neoplasia. The levels of Ezrin protein expression in invasive cervical carcinoma stageⅡ,Ⅲwere significantly higher than the group of normal cervix,and the levels of Ezrin protein expression in invasive cervical carcinoma stageⅢwere significantly higher than the group of cervical intraepithelial neoplasia.It indicates that RT-PCR method is more sensitive to detect Ezrin expression, Ezrin mRNA is more likely to become a marker to detect the progression of cervical cancer2. Ezrin mRNA and protein expression levels in cervical cancer tissues were statistically significant in the depth of cervical stromal invasion and lymphnode metastasis in cervical carcinoma surgery patients (P<0.05). Ezrin may have an important role in predicting cervical lymphnode metastasis. Ezrin can be used as a prognostic indicator for the judgment of cervical cancer. Ezrin may become a new target to treat tumor metastasis in the future.3. Ezrin protein and Ezrin mRNA expressions were positive correlation. It indicates that their expressions are parallel relationship.4. Ezrin mRNA and protein expression levels in cervical cancer tissues were not statistically significant in different pathological grade,pathological type, tumor size and age (P>0.05).
