PartⅠThe relationship between VEGF,PEDF and inner blood-retinal barrierPURPOSETo observe the relationship between VEGF,PEDF and inner blood-retinal barrier in diabetic rat.METHODS(1)Take the normal male Wistar rats as normal contral group,diabetic models induced with streptozotocin(STZ)were divided into four groups:4-weeks diabetic group,4-weeks PBS group,4-weeks Avastin group and 4-weeks PEDF group.(2)intravitreal injection:PBS was injected into the right eye of PBS group at 2μL/eye,2μL Avastin to Avastin group,2μL recombinant PEDF to PEDF group,diabetic and normal group were pseudo-injection.(3)The retinal vascular permeability was determined by Evans Blue.(4)The protein expressions of VEGF and PEDF in retina were determined by immunohistochemistryRESULTS(1)Retinal Evans blue leakage of eyes from 4-weeks diabetic group were 2.68-fold hitcher than that of normal group,the leakage from the Avastin-injected and PEDF-injected eye were 0.36-fold and 0.39-fold than self control eye respectively,but have no significant difference with normal group.(2)expression of VEGF in diabetic group were higher than those in normal group,PEDF were lower.(3)expression of VEGF in Avastin-injected eye were lower than those in self control eye and diabetic group,but PEDF have no significant difference.(4)expression of VEGF in PEDF-injected eye were lower than those in self control eye and diabetic group,but PEDF were higher than them.CONCLUSIONS(1)Inner blood-retinal barrier breakdown can occur at the early stage of diabete,it followed by the upregulation of VEGF,downregulation of PEDF.(2)inhibit the VEGF or upregulation PEDF to keep the balance of PEDF/VEGF can reduce retinal vascular permeability. PartⅡThe effect and mechanism of Fufang XueShuan Tong capsule on diabetic macular edemaPURPOSETo observe the effect and mechanism of Fufang XueShuan Tong(FXST) on diabetic macular edema,we investigated the effect of FXST on the inner blood-retinal barrier.METHODSTake the normal male Wistar rats as normal contral group(by the way of given Sodium chloride from esophagus to stomack,1 time/day,experimental period was 4 weeks );diabetic models induced with streptozotocin were divided into three groups:diabetic contral group(same treatment as the normal group),FXST-M treatment group(with FXST at dose 900mg/kg) and FXST-H treatment group(at dose 1800mg/kg).Measurement of blood-retinal barrier breakdown using Evans Blue.The protein expressions and mRNA of VEGF and PEDF in retina were determined by immunohistochemistry and RT-PCR,respectively.RESULTS(1)Retinal Evans blue leakage of eyes from diabetic contral group was 2.68-fold,2.60-fold and 2.73-fold higher(P<0.05) than that of normal group,FXST-H and FXST-M group,respectively.the leakage from FXST-H and FXST-M group had no significant difference with normal group(P>0.05).(2)The protein and mRNA of retinal VEGF in diabetic control group were higher than in normal group(P<0.05),PEDF was lower than normal group(P<0.05).The levels of VEGF in FXST-M and FXST-H group were lower than diabetic control group(P<0.05),but had no significant difference with normal group,PEDF were lower than normal group but no significant difference with diabetic control group.CONCLUSIONSFXST stabilized the inner blood-retinal barrier and had beneficial effect on diabetic macular edema in association with regulation of VEGF expression in the retina.
