ObjectiveTo evaluate the biological effects of allicin on the proliferation and protein synthesis,as well as ultrastructural changes,of in vitro cultured human periodontal ligament cells(HPDLCs) after stimu-lating by differrent concentration allicin,and to examine protecting/habiting action of allicin to HPDLCs,preliminarily explore possible mechanism of allicin to HPDLCs,providing experimental and theorial basis of a new intracanal medication for root canal therapy.MethodsCultured in vitro HPDLCs,then were stimulatd by different concentration Allicin such as 0mg/l(control group),10-2 mg/l,10-1 mg/l, 1mg/l,10 mg/l,102mg/l,103mg/l.And then we used the MTT,coomasie brilliant blue staining assay to detect the expression of cell proliferation and total protein contents in these groups.The transmission electron microscope(TEM) method was applied to observe cells' ultrastructure.Results(1) The concentration of allicin in the 10mg/l,102mg/l,103mg/l significantly inhibited the proliferation of human periodontal ligament cells,but no significant effect in the 10-2mg/l,10-1mg/l,100mg/l and the effect of 103mg/l allicin was the lagerest.The role of allicin showed a clear concentration-dependent effect,the inhibitory concentration 50% approximately 22mg/l.(2) The concentration of allicin in the 10mg/l,102mg/l,103mg/l signi-ficantly inhibited the total protein content of HPDLCs,and the concentration of 103mg/l was at the maximum effect,(ANOVA and S-N-K test,P<0.05)(3) Different concentrations of allicin have different degrees of ultrastructural damage on human periodontal ligament,and 10mg/l, 102mg/l,103mg/l groups showed obvious injury.Conclusion(1) Allicin had a inhibition of cell proliferation and protein synthesis on HPDLCs,and showed a clear concentration-dependent effect,the inhibitory concentration 50%approximately 22mg/l.(2) Allicin had an inhibition of varying degrees of ultrastructural damage.
