Objective Investigate the extracting technology of the Arnebiae euchroma by means of systematic solvent extraction,and study on the valid part that the thing have antibiosis function,what extracted from the Arnebiae euchroma by means of systematic solvent extraction,to provide a scientific basis for antibiosis.Methods Adopt the systematic solvent extraction to extract one by one in order,the optimum extraction process was selected with the orthogonal design method and singal factor design method,divide the segment system kind,the extraction infuses the stomach to the big rat and the small rat,after stopping the medicine several days dissect the animal,record the number of nidation inside the cape order,the living embryo and still-born babies number. The X~2 examination was used to compare pregnancy rate of control group with medication whether have significant deviation or not. The T examination was used to compare the number of nidation,the living embryo and still-born babies number in control group with medication group whether have significant deviation or not. Result (1) Extraction process:the Arnebiae euchroma in 50℃,drying 3h,screen off on the 3rd ,from medicine powder,according to law percolation operation,increase petroleum ether adequate wetting and installing extinguishers,exhaust,soaked 12 hours to contrast rate of cold infiltration shenlu. Percolation fluid collection,low recovery of petroleum ether percolation condensed to liquid syrup-and low-temperature vacuum drying extract into standby;Plus residue with ethyl acetate,ethanol cold infiltration shenlu,extract from the reserve;Finally residue with boiling water twice every time an hour,plus 10 times the amount of water combined boiling liquid,low-enriched dry extract standby.(2)Adopt the systematic solvent extraction was used to separate extracts from the Arnebiae euchroma, the different extraction to gain the plain content dissimilarity of L-Shikonin,the content of the L-Shikonin of petroleum ether,the alcohol extraction was 1.38%,0.62%,0.01% respectively,the water extraction can not measure the L-Shikonin.(3) The Arnebiae euchroma petroleum ether, ethyl acetate, ethanol, water extract maximum tolerance respectively: 2g/kg,12.5g/kg,19.5g/kg,23.4g/kg.(4)The pharmacodynamic experiment indicated:the Arnebiae euchroma petroleum ether extraction had the anti-implantation and the antiearly pregnancy effect,but the other extraction had not.(5) The verification experiment result express,the anti-implantation,the antiearly pregnancy of big rat was in accordance with the small rat of the experiment result early. Conclusion Optimum use of the system solvent extraction process,using the Arnebiae euchroma nature of the chemical composition differences the Arnebiae euchroma extract the various ingredients,use modern anti-fertility pharmacodynamics experiment choice the Arnebiae euchroma different solvent extraction from the effective anti-fertility,this will be the Arnebiae euchroma effective anti-fertility effect on the composition and structure of the breakthrough.Meanwhile,the efficacy results showed the Arnebiae euchroma petroleum ether extract with anti-implantation,the pregnancy-,whether preimplantation early pregnancy or delivery can terminate pregnancy.the Arnebiae euchroma anti-fertility mechanism may be soft in the Arnebiae euchroma the role of gonadotropin-inhibiting ovulation the egg in the process and the impact of the luteal secretion inhibitors or with the nature of proliferation,PTKs,it is anti-animal and anti-implantation pregnancy. For further separation monomer,the Arnebiae euchroma rationalize anti-fertility effect with the relationship between the development of anti-fertility medicine,have some far-reaching significance.
