As our national medicine, traditional Chinese medicine do have played a very important role during the process of struggle with disease. But with the progress of the era, traditional Chinese medicine have gradually lost its advantages. So it is very important of the modernization of the traditional Chinese medicine to keep it in an important position in the national history and culture. However, the active ingredient is the basic material of a traditional Chinese medicine to take effect, so a in-depth study of the active ingredient is the core step in the modernization of the traditional Chinese medicine. In this paper, the extraction, separation and purification of the coumarin from Angelica dahurica have been studied respectively. And the novel method in this paper can provid a prospect for the industrialization of the separation and purification of coumarin from Angelica dahurica. The main researches are as follows:1. Angelica dahurica was extracted by supercritical CO2 (SC-CO2). Through orthogonal test (L934), the key parameters and the best technological conditions during the supercritical fluid extraction (SFE) can be determined. And in this paper, coumarins were set as the target ingredient to judge the quality of the product.2. In this part, we use scientific method to determine the solubility of imperatorin in five different solvents. All the data can not only contribute to the accurate design of the separation and purification process, but also to the research of dosage form and pharmacological action.3. In this process, imperatorin and isoimperatorin were set as the target componds. The solvent extraction was used as the aeparation procedure. Thouth a series of experiments, the exteaction solvent and the best extraction conditions can be obtained.4. In this part, though choosing the appropriate crystallization solvent and condition, the crystals of the coumarins with high purity can be obtained.
