Objective:To investigate the expression of ET-1 and NOS in gingival tissue of chronic periodontitis patients,gingivitis suffers and periodontally healthy controls,to investigate the correlation between clinical parameters and laboratory data,and to explore the possible mechanisms of inflammatory periodontal diseases and find new therapeutic pathways.Method:Immunohistochemistry and technology of computer picture disposal were used to detected the expression of ET-1 and NOS in tweenty samples of gingival tissue from clinical healthy individuals, respectively;tweenty samples of gingival tissue from clinical healthy individuals,the gingivitis and the periodontitis respectively.Result:1.Expression of ET-1 protein:①Positive immunostaining with the antibody was observed in the cytoplasm of the epithelium and endothelium cells.②The positive expression rates of ET-1 in gingival tissue of normal people,the gingivitis and periodontitis were 50.00%(10/20),85.00%(17/20) and 90%(18/20);the positive expression rate of ET-1 in gingival tissues of gingivitis and periodontitis were higher than that of normal people.③The average photodensity of ET-1 in gingival tissue of normal people,the gingivitis and periodontitis were 0.26±0.02,0.31±0.07 and 0.37±0.09;The average photodensities in gingival tissues of gingivitis and periodontitis were higher than that of normal people, and that of periodontitis was higher than gingivitis.④In gingivitis group,a significant positive correlation was found between the photodensity of ET-1 in the gingival tissue and traditional periodontal parameters PD,GI and SBI.⑤In periodontitis group,a positive correlation was existed between the photodensity of ET-1 and PD,GI,SBI and AL.2.Expression of NOS protein:①Positive immunostaining with the antibody was observed in the cytoplasm of the epithelium and endothelium cells,and some in the inflammatory cell of the connective tissue.②The positive expression rates of NOS in gingivai tissue of normal people,the gingivitis and periodontitis were 20.00%(4/20),55.00%(11/20) and 60.00%(12/20);the positive expression rate of NOS in gingival tissues of gingivitis and periodontitis were higher than that of normal people.③The average photodensity of NOS in gingival tissue of normal people,the gingivitis and periodontitis were 0.18±0.01,0.26±0.04 and 0.22±0.06;The average photodensities in gingival tissues of gingivitis and periodontitis were higher than that of normal people, and that of periodontitis was lower than gingivitis.④In gingivitis group,a significant positive correlation was found between the photodensity of NOS in the gingival tissue and traditional periodontal parameters PD,GI and SBI.⑤In periodontitis group,a negative correlation was existed between the photodensity of NOS and PD,AL.3The correlation relationship of the expression of ET-1 and NOS in gingival tissue:①In gingivitis group,a positive correlation was found between the expression of ET-1 and NOS in gingival tissue,P<0.05.②In periodontitis group,a negative correlation was found between the expression of ET-1 and NOS in gingival tissue,P<0.05.Conclution:The expression of ET-1 and NOS in gingival tissue may be associated with the initiation and progression of periodontal diseases.The expression of ET-1 in gingival tissue can be statistically related to the cinical paraments at inflammatory sites in patients with gingivitis and periodontitis,which suggests that changes of the ET-1 partially reflect the results of destruction in periodontal tissue.The expression of NOS in gingivai tissue can be statistically related to the cinical paraments at inflammatory sites in patients with gingivitis,and the attachment loss as well as pocket depth,which suggest that changes of the NOS reflect the degree of inflammatory destruction in early stage.There exist the disbalance of ET-1 and NOS in gingival tissue of periodontal disease,the interaction among them may plays an important role in the process of periodontal disease.
