Objective:To investigate the expression of EphB4 and EphrinB2 in colorectal carcinoma and their relation with angiogenesis,and compared it with that of precancerous lesion and that of normal colorectal tissues,to find out the relationship between EphB4 receptor and its ligand EphrinB2 with clinical pathological factors.Method:Immunohistochemistry was applied to examine the expression of EphB4 receptor and its ligand EphrinB2 in 63 cases of human colorectal carcinoma, 30 cases of colorectal adenoma,25 cases of colorectal polyp and 25 cases of normal colorectal tissues.Results were compared with clinical pathologic parameter such as lymphatic metastasis and clinical stage.Result:①Positive immunostaining with the antibody of EphB4 receptor and its ligand EphrinB2 was observed in the nuclear and cytoplasma of the carcinoma and precancerous lesion.②There was a significant positive correlation between carcinoma and precancerous lesion specimens with normal mucosa specimens(p<0.05).③The expression of EphB4 and EphrinB2 protein had a significant positive correlation with differentiation grade,infiltration,lymph node metastases and clinical stage(p<0.05). There was no relation to the patients' age,sex,tumor size and location(p>0.05).④EphB4 and EphrinB2 may be closely associated with angiogenesis(p<0.05).Conclution:The EphB4 and EphrinB2 protein expression increase may be involved in development of colorectal carcinoma and play important roles in the tumorigenesis and progression of colorectal carcinoma.
