Objective: To investigate the effects of GnRHa and Gestrinone on growth, proliferation and apoptosis in isolated ectopic and eutopic endometrial stroma cells from women with endometriosis, as well as the expression of MMP-3 induced by GnRHa and Gestrinone in vitro.Methods:The ectopic and eutopic endometrial stromal cells were isolated and cultured in vitro. Then they were identified by Vimentin immunocytochemistry staining. Both the ectopic and eutopic endometrial stromal cells were exposed to LA and Gestrinone at different concentrations for 0-96h. The proliferation and morphological changes were observed by inverted microscope. The growth and inhibition of ectopic and eutopic stromal cells were measured by MTT assay. The apoptosis and cell cycle distribution induced by different concentration of LA and Gestrinone on ectopic and eutopic stromal cells were assayed by flow cytometric analysis. The expression of MMP-3 induced by LA and Gestrinone was analyzed by immunocytochemistry.Results:The purity of cultured ectopic and eutopic endometrial cells was above 95% identified by immunocytochemistry staining. The growth and proliferation of ectopic and eutopic endometrial cells were inhibited in a dose-and time-dependent manner by LA and Gestrinone at different concentrations. LA and Gestrinone could obviously block the cell cycle of ectopic and eutopic endometrial stroma cells with the number of cells increased at G1 stage and decreased at S stage; The apoptotic rate of endometrial stroma cells were increased significantly comparing with that of the control group (p<0.05); The expressions of MMP-3 induced by GnRHa and Gestrinone were obviously decreased (p<0.05), But the inhibition of LA and Gestrinone to the eutopic endometrial stroma cells was pooer than to ectopic endometrial stroma cells. In contrast, the apoptosis was enhanced more signifieantly and the expressions of MMP-3 were decreased more obviously by GnRHa than that of Gestrinone (p<0.05).Conclusion: 1,GnRHa and Gestrinone could significantly inhibit the proliferation and enhance the apoptosis of ectopic and eutopic endometrial stromal cells of endometriosis, the former was more stronger.2,GnRHa and Gestrinone could obviously decrease the expression of MMP-3 with the former stronger.3,The actions above were more stronger to ectopic endometrial stroma cells than to eutopic endometrial stroma cells.
