Microglia Isolation Method And Cell Identification

Objective: To establish a method of improved efficient primary mouse microglia isolation and its purification. Methods: Primary microglia culture was modified based on the classicmicroglia culture method of McCarthy's. Theisolation of the microglia from mixed cultured cells was by hand. Microglial cells were identified by immunohistochemistry using mouse anti rat CD11b monoclonal antibody(0X42) . Results : High purity of Primary microglial cell was obtained. Conclusion: The purity of microglia by the method of mildtrypsinization was equal to that of M cCarthy S. but the efficiency of microglia isolation is improved.
