Objective: To investigate the expression of Slit2 and its correlation with the microvessel density in human hypopharyngeal squamous cell carcinomas, and to analyze their influences to the biological behaviors of human hypopharyngeal squamous cell carcinomas.Method: The expression of Slit2 protein and CD34-labeled microvessel density (MVD) were measured by immunohistochemical staining (Envision). The expressions of Slit2 mRNA in 24 cases of fresh tissues were measured by reverse-transcript polymerase chain reaction. The relations between the above expressions and the clinical features of these patients were analyzed by statistical methods.Results The positive rates of Slit2 in cancerous tissues and Para-cancer tissues were respectively 65.2% and 40.5% and the difference between them had statistical significance (X~2=6.329, P<0.05).The Slit2 mRNA expressions between them also had statistical significance (t=8.230, P<0.05). The expression rate of Slit2 and the enumerations of MVD were significantly associated with tumor size, TNM stage and lymph node metastasis. The enumerations of MVD were high in Slit2-positive tissues, and the difference had statistical significance (t=2.273, P<0.05) . The expression of Slit2 had positive correlation with the MVD (r_s=0.430, P<0.05) .Conclusion: 1. The expression of Slit2 and the enumerations of MVD were significantly higher in hypopharyngeal squamous cell carcinomas than that in normal mucosa. The relation between them in cancerous tissues was positive.2. The expression of Slit2 and the enumerations of MVD had significant relations with the tumors' clinical features, such as invasion and lymph node metastasis. The mechanism might be Slit2-mediated new vessels formation, which affected the tumors' biological behaviors.3. The expression intensity of Slit2 and the enumerations of MVD can be used as an important index for the capacity of growth, invasion and metastasis in the hypopharyngeal squamous cell carcinomas.
