Conjugated linoleic acid (CLA) is a generic term of positional and structural isomers of linoleic acid. CLA has been reported to have diverse physiological functions like anti-carcinogenicity, blood lipid regulation activity, anti-atheroscleroticity, immunoregulation effects, as well as body fat reduction effects. Structured lipids containing functional fatty acids, due to their special nutrition and functions, are attracting more and more attention. The preparation, purification and function of the structured lipid containing CLA (CLA-SL) were studied in the present study.CLA-SL was preparated by enzymatic acidolysis of CLA and sunflower seed oil in the solvent-free system, with the optimum reaction conditions obtained though factorial experiments: water content 1%, ratio of CLA to sunflower seed oil 3.55:1, enzyme load 13.7% of substrate, reaction temperature 55oC, and reaction time 37h. Under these conditions, CLA-SL was obtained with a total CLA incorporation rate of 17.1% and a sn-2 position CLA incorporation rate of 2.89%.CLA-SL was subjected to molecular distillation for purification, with the optimum coditions determined: preheating temperature 80oC, feed materiel quantity 2mL/min, heating temperature 180oC, scraper rotation rate 100r/min, condensing temperature 25oC and vaccum pressure 1Pa. Moreover, the physiochemical properties of CLA-SL were determined: color Y20R2.2, acid value 0.23mgKOH/g, peroxide value 3.15mmol/kg, iodine value 131gI/100g and saponification value 192mgKOH/g.Animal test in mice indicated that CLA-SL can reduce tricylglycerol concentration of serum and increase HDL-C concentration significantly. Moreover, CLA-SL may have anti-atherosclerotic effect since it can reduce AI value and increasing AII value significantly. Furthermore, CLA-SL can reduce body fat and increase body water and protein significantly without substantially reducing body weight.
