β2-microglobulin(β2-MG)was used to tests of renal function and considered as an endogenous substance that was more sensitive than serum creatinine. In recent years, it could be in widespread use clinically. This needs to set up a convenient, accurate and fast new detection method. In this study, using McAb optimize the double antibody sandwich method of ELISA and build a fast new method onβ2-MG detection. It can detect levels ofβ2-MG in serum of hepatopaths, nephrotics, diabetics and health adults by this method, and probe values of changing serumβ2-MG content in clinical diagnosis.The results showed that, the optimum experimental condition of detecting serumβ2-MG by the double antibody sandwich method of ELISA was that diluted antibody 6E5(4.2mg/ml) to 1:800, diluted ELIAS antibody 7H5-HRP to 1:150, not diluted samples of serumβ2-MG , putted ELIAS plate in 37℃for 2 hour then in 4℃for 12 hour after coating ,and confined for 2 hour. By this method, the serumβ2-MG content of 294 hepatopaths, 75 nephrotics, 63 diabetics and 113 health adults were detected by this method. The results by statistical analysis showed that : There was remarkable significance of difference between the serumβ2-MG level of liver disease groups and normal group (P<0.01). Significantly positive correlations were observed between the serumβ2-MG levels and ALT/AST; There was remarkable significance of difference between the serumβ2-MG level of nephronia patients and normal group (P<0.001). And significantly positive correlations were observed between the serumβ2-MG levels and the urineβ2-MG levels; There was significance of difference between the serumβ2-MG level of diabetic patients and normal group (P<0.05), and the positive rate of the serumβ2-MG (74.6%) was higher than the positive rate of the urineβ2-MG (55.6%). In liver disease and nephronia, the positive rate of hepatic carcinoma and renal carcinoma were both highest. There were 79.2% and 90.3%. These studies proved that was the double antibody sandwich method of ELISA not only a convenient, fast and accurate method of detecting serumβ2-MG content, but the detecting result of serumβ2-MG by this method can serve as a guideline for diagnoses of liver disease, nephronia, diabetic nephropathy and phymatosis in clinical diagnosis. And it was of reference value for judging extent of tissue damage and the prognosis.
