The trend in cosmetic surgery over past years has been a shift towards minimally invasive procedures with minimized recuperation intervals. Radiofrequency treatment for wrinkles has been recently utilized for noninvasive facial rejuvenation. However, the shortage of knowledge in the mechanisms restrained the development and the further use of radiofrequency treatment for wrinkles. Hence, we observed Radiofrequency treatment for wrinkles systematically and did some animal and cell experiments to explain the partial mechanism and facilite the practice.Objectives:(1) To evaluate the efficacy of the radiofrequency device which was used.(2) To evaluate the effect of radiofrequency current on the cell proliferation and collagen hyperplasy in cultured fibroblasts by applying radiofrequency current to the cell cultures.(3) To evaluate the histologic change in the dermis and the change of the rate of typeⅢand typeⅠcollagen by treating the rabbits with the radiofrequency device.Methods:(1) Choosing 20 patients with wrinkles in the face, all of them receive a course of radiofrequency treatment. Every patient was treated for five times at an interval of 10 days, Patient feedback about heat sensation is a valid and preferred method for selecting the optimal amount of energy to use. At the end of the course the effect was recorded.(2) A electric device was designed for applying radiofrequency current to the cell cultures. Draw the grouth curve of fibroblast, Observe the changes of cell circle by the means of flowing cytometry, Proliferating cell nuclear antigen (PCNA) was examined by immunohistochemstry, then, evaluate the effect of radiofrequency current on the cell proliferation; The quantitative assay of collagen protein was detect by incorporated to evaluate the effect on the collagen hyperplasy. (3) To observe the effect of radiofrequency current on the dermis, ten rabbit were treated by radiofrequency, and the histologic change in the dermis were observed by HE staining and Sirius red staining.Results:(1) We observed the curative effect of the RF current was conspicuous, Radiofrequency had a great curative effect to the low-grade wrinkles, but a little curative effect to the serious wrinkles, otherwise, the medium wrinkles can be improved obviously.(2) Compared to the control group, there were noticeable effect on the cell population of fibroblasts treated by RF current.(3) By culturing fibroblasts 24 hours after the treatment with RF current, compared with the distribution in cell circle in the RF groups and the control group, it was showed that the percentage of the DNA level in the S and G2-M phase was higher significantly than in the control group (P<0.05), while in the percentage of G1 phase was lower than G2-M phase(P<0.05).(4) By culturing fibroblasts 24 hours after the treatment with RF current,Proliferating cell nuclear antigen was examined by immunohistochemstry , Compared to the control group, Proliferating cell nuclear antigen expression of fibroblasts was increased and increase with the increment of the energy(P<0.05).(5) To detect the collagen synthesis of fibroblast with the technology of incorporation 3H- pyrrolidinecarboxylic acid. By culturing fibroblasts 24 hours after the treatment with RF current, the quantitative assay of collagen protain was were increased (p<0.05), There is noticeable change between each group. The quantity of incorporation is 1226.62±165.74(Cpm)in the control group, but it is 3202.95±205.29(Cpm)in the 20Vpp group. So the radiofrequency current can increase the incorporation of the 3H- pyrrolidinecarboxylic acid in human fibroblasts.(6) After RF treatment for about 1 month , the fibers in the dermis appear more compact.(7) After RF treatment for about 1 month, the quantity of the typeⅢ(red) and typeⅠ(green) collagen were both increased, the fibers in the dermis appear more compact. and the rate of typeⅢand typeⅠcollagen was increased.Conclusion: (1) The clinical effective rate of the RF current to the wrinkles was 95%.(2) The radiofrequency current can induce the cell proliferation and increase the synthesis and secretion of collagen of fibroblasts.(3) The radiofrequency current can increase the quantity of collagen in the dermis,and increase the rate of typeⅢand typeⅠcollagen。...
