Objective:To study the protection role of recombinant human erythropoietin(rhEPO) on the rats with acute kidney damage induced by the open abdominal wound combined with artificial seawater immersion.Methods:On the bases of 72 rats preliminary experiment,the 91 healthy male Wistar rats (clean grade,body weight 247.53±28.97g) were randomized divided into 6 groups.The control group 16 rats,only anesthetized without open abdominal wound nor artificial seawater immersion,8 rats for getting blood and kidney samples at 3 hours,8 rats for getting blood and kidney samples at 6 hours.Other 5groups,there were 15 rats for each group.The EPO pretreatment group rats were given 1000U/kg EPO before they received open abdominal wound and artificial seawater immersion for 3 hours,then got the blood and kidney samples. The experimental group rats received open abdominal wound and artificial seawater immersion for 3 hours,then got the blood and kidney samples.The small dose EPO group rats were given 1000U/kg EPO after they received open abdominal wound and artificial seawater immersion for 3 hours,then were put in room temperature(30℃) for other 3 hours, and then got the blood and kidney samples.The large dose EPO therapy group rats were given 2500U/kg EPO after they received open abdominal wound and artificial seawater immersion for 3 hours,then were put in room temperature(30℃) for other 3 hours,and then got the blood and kidney samples.The later observation experimental group rats received open abdominal wound and artificial seawater immersion for 3 hours,then were put in room temperature(30℃) for other 3 hours,and then got the blood and kidney samples.The open abdominal wound combined with artificial seawater immersion(22℃) were carried on rats except control group to induce the acute kidney injury model.The blood creatinine,urea nitrogen,creatine kinase,creatine kinase isoenzyme,TNF-α,IL-6, C3a,c-reactive protein,kidney tissue homogenate SOD and pathologic changes of kidneys were studied.The data were showed as x±s,the statistics were carried with SPSS11.5.The P value<0.05 was considered as significant different.Results:Compared with the control group,other 5 group rats occurred acute kidney injury after the open abdominal wound combined with artificial seawater immersion for 3 hrs,with significant serum creatinine and urea nitrogen increasing,electrolyte disturbances, acidosis,but could be survival.Pretreatment group rats serum creatinine and urea nitrogen, creatine kinase,creatine kinase isoenzyme,TNF-α,IL-6,C3a,c-reactive protein,kidney tissue homogenate SOD and pathologic changes of kidneys were better than thoses of experimental group(P<0.05).Both small dose and large dose EPO therapy groups had better income with serum creatinine and urea nitrogen,creatine kinase,creatine kinase isoenzyme,TNF-α,IL-6,C3a,c-reactive protein,kidney tissue homogenate SOD and pathologic changes of kidneys than the later observation experimental group(P<0.05). There were not obvious different between small dose group and large dose group(P>0.05).Conclusion:1.We made the open abdominal wound plus artificial seawater immersion induced rat acute kidney injury model successfully.2.EPO could relieve inflammatory reaction,enhance antioxidant enzyme,and lighten acute kidney injury of rats induced by open abdominal wound combined with artificial seawater immersion.
