Objective: To establish a diabetic rat model,to give low, medium and high intensity exercise three types of intervention, observation of three different intensity of exercise on blood glucose in diabetic rats and other related parameters of the effects of exercise therapy of diabetes to explore the best exercise intensity and its mechanism with a view to formulate diabetes exercise prescription for reference.Methods: Wister male rats 70, will be the end of intravenous normal saline 8 rats as normal control group (C); other 62 as the DM modeling group, modeling group separately by 50mg/kg, divided into two four times the tail vein injection of alloxan, the establishment of rat model of DM, the rats into a mode selected randomly divided into 4 groups, that is quiet DM group (DQ), DM of low-intensity exercise intervention group (DSL), DM in the intensity of exercise intervention Group (DSM), DM high-intensity exercise intervention group (DSH); determine the animal training methods, namely, low-intensity (15min / m), Medium Intensity (27min / m) and High Intensity (33min /m), training every day 25 min, five days a week.Take three different exercise intensity on the DSL groups, respectively, DSM Group, DG intervention group were observed before and after interven- tion in weight,fasting blood glucose (FBG) and post-intervention blood lipid, serum insulin (Ins), glycosylated serum protein (GSP) , serum NO, NOS content and islet B-cell function index (HBCI) changes.Results: To take the tail vein injection (50mg/kg, twice) alloxan approach, can successfully set up in line with the characteristics of human diabetic Wister rat model. The experimental results show that: (1) the adoption of low-intensity, medium-intensity, high intensity in three different forms of exercise intervention, the exercise group the value of fasting blood glucose in diabetic rats (FBG ), blood lipids (TC, TG, LDL-C, HDL-C), glycosylated serum protein (GSP), serum NO and NOS were significantly lower than the quiet group of diabetic (DQ), has a very significant difference (P <0.01); while the serum insulin (Ins), pancreatic B-cell function index (HBCI) higher than the quiet group of diabetic (DQ), has a very significant difference (P <0.01). (2) of the three diabetic exercise group compared with medium-intensity exercise group (DSM) and high intensity exercise group (DSH) of the fasting blood glucose value (FBG), respectively, lower than the low-intensity exercise group (DSL), and there is a significant difference (P<0.05), DSM group is lower than the fasting blood glucose DSH group, but there is no difference (P>0.05); DSM Serum insulin (Ins) levels higher than the DSL group and, with significant differences (P <0.05), DSH is higher than the level of Ins Group DSL Group, DSM Group Ins level higher than the DSH group, but not with differences (P>0.05) ; DSM group and the DSH group ofβ-cell function index (HBCI) are significantly higher than the DSL group, have a very significant difference (P <0.01), DSM Group HBCI than DSH group, there is no difference (P>0.05); in triglyceride (TC), cholesterol (TG), low density lipoprotein (LDL-C) on the comparison, DSM group minimum, DSH Group followed, DSL group the highest, and they do not have differences (P>0.05),high density lipoprotein(HDL-C)DSM highest,than DSH Group followed, DSL group the lowest, and not statistically significant (P>0.05); at glycosylated serum protein (GSP) , serum NO level and NOS on the comparison, DSM group have less than the DSL group, significant differences (P <0.05), DSM group than in DSH group, DSH group than in DSL group differences did not exist (P> 0.05).Conclusion: (1) Through Wister rats 50mg/kg twice by tail vein injection of alloxan, successful set up a diabetic rat model (model a success rate of 59%) for diabetes research laboratory premise conditions. (2) low, middle and high three different intensity treadmill exercise on disease prevention and treatment of diabetic rats have an active role. (3) medium-intensity treadmill exercise (27m/min) of diabetic rats blood glucose, blood lipids, lowering of glycosylated serum protein and serum insulin levels, isletβ-cell function improve the effect of the most obvious, is superior to low, high-intensity exercise.
