| [Objective]To study the effects of Naofucong(NFC) on Morris water maze test and expression of NF-kB,GFAP in hippocampus of STZ-induced diabetic rats.[Methods]SET-induced diabetic rats(single intraperitoneal injection) were randomly divided into 5 groups including nomal group,model group,low-dose NFC group,middle-dose NFC group,high-dose NFC group.All rats were given intragastric administration for 8 weeks,the normal and model groups were treated with distilled water.Cognitive function was tested in a Morris water maze before STZ injection and 8 weeks after intragastric administration. Expression of NF-kB,GFAP was tested ex vivo,in hippocampal slices by SABC immunohistochemical method.[Results]1.Morris water maze test:The time spent in target quadrant by rats with 8 week diabetes was significantly longer than the time spent by control animals.The time spent in target quadrant by rats in all the treated group was significantly shorter than the time spent by diabetic rats.Spearman correlation analysis showed there was negative correlation between the time spent in target quadrant by treated rats and the dose of Naofucong.2.Results of immunohistochemical in hippocampal slices:(1) Expression of NF-kB in CAlof hippocampus:Expression of NF-kB in diabetic and treated rats was significantly higher than in normal rats. Expression of NF-kB in treated rats was significantly lower than in diabetic rats.Spearman correlation analysis showed there was negative correlation between the expression of NF-kB in treated rats and the dose of Naofucong. (2) Expression of GFAP in hippocampus:The integrated option density of GFAP expression in STZ-DM,low-dose NFC group and middle-dose NFC group rats was much lower than the normal rats.And the levels of GFAP in middle and high dose NFC group rats increased notably compared with model group. Spearman correlation analysis showed there was positive correlation between the expression of GFAP in treated rats and the dose of Naofucong.[Conclusion]Diabetic rats spent more time in target quadrant in Morris water maze test 8 weeks after SZT injection,which demonstrated the models of cognitive impairment were established.Expression of NF-kB in diabetic rats was significantly higher than in normal rats,which suggested the chronic inflammation of hippocampus was induced.Expression of GFAP in diabetic rats was much lower than the normal rats which suggested the missing of synaptic transmission,oligotrophic neuron and the missing of differentiation in neuron stem cell.The cognitive impairment of diabetic rats was related to the decrease of GFAP and increase NF-kB expression in hippocampus. Traditional Chinese Medicine Naofucong could improve cognitive function, up-regulate the expression of GFAP and decrease the expression of NF-kB in hippocampus.The high dose of NFC showed the best effectiveness. |
