| [Background]:When the diameter of matter less than 100nm,its physical and chemical nature will be changed.And because it has more powerful surface activity,nano-particals would bring injure to living cells.Following the application of Nano-magnetic Fluid in medical science, its safety become more and more important.But at present the study for the toxicity of nano-particals is not sufficient,especially about its lung and heart toxicity.We know,that have confined zhe use of nano-materials.This article is just about the toxicity of Fe203 nano-particals.[Objective]:To inject zhe drug with different doses into mice audal vein.And 20-days latter we investigate its oxidative effects,which include the change of enzyme,the change of pathology appearance and the change of apoptosis index(AI).[Method]s:We use L-glutamic acid as surfactant to prepare Fe203 Nano-magnetic Fluid and inject the drug to make mice poisoned.After we collect blood from fossa orbitalis,and kill the mice.To select lungs and hearts of test mice and use parts of lungs for evaluating wet and dry ratio. The other specimens are prepared tissue bomogenate or cut sheets,in order to determine enzymes correlated with oxidative damage,pathology characteristics,and effect of nano-particals upon apoptosis,which is done by TUNEL staining.[Results]:After the treatment,the LDH in blood serum increased(P<0.01);the MDA,MPO also increased in hearts and lungs,and the activity of SOD decreased(P<0.01).But zhe enzymology changes of heart are not significant when we inject the low dose.We also find that the increase of MPO activity test mice's lungs coincide with its pathology change.Cell apoptosis in experimental animals' lungs have notable changes V.S control group(P<0.05,P<0.01).But about cardiac muscle only in high dose group apoptosis have statistical significance compared with control group(P<0.05).[Conclusion]:The Fe2O3 Nano-magnetic Fluid,using L-glutamic acid as surfactant can cause obviously oxidative damages to test animals' lungs and cardiac muscle.Nano-magnetic Fluid make some materials correlated with oxidative damage significantly changed.And that also influenced cell apoptosis,especially for lung tissues. |
