ObjectiveTo investigate the relationship between NF-κB expression and neuron apoptosis after cerebral ischemia reperfusion injury in rats. And through the experiment to investigate whether atorvastatin-calcium has impact on the expression of NF-κB levels and the impact of neuronal apoptosis in the brain tissue after ischemia/reperfusion (I/R) injury.MethodsOne hundred and five healthy male Wistar rats,supplied from animal experiment center of Lanzhou university,were devided into shamoperation(Sham)group,ischemia/reperfusion(I/R)group,atorvastatin-calcium intervention group. Atorvastatin-calcium intervention groups were given Atorvastatin calcium at doses of 5mg/kg.The drugs were given once a day and consecutively for twenty days before operation.The rats were chosen to establish focal cerebral ischemia reperfusion models with middle cerebral artery thread embolism method. After animal model were successfully established,neurological deficits were measured,the scores are all in 2-3,assuming to enter the groups after the rats were awake by means of levo circling when crawling,left anterior limb adducting and flexion when lifting tails,besides subarachnoid hemorrhage.After the operation,the rats were back to cages,eating food and water freely. Detecting contents consists of four aspects:1.Each group was undertook after 2 h ischemia,3 h reperfusion injury,to observe Atorvastatin calcium's action to rats' nervous function after cerebral ischemical reperfusion injury.2. Measure rats'cerebral infarction volume: To observe Atorvastatin calcium's effect to rats'cerebral infarction volume.3. Observe Atorvastatin calcium's effect to rats'nerve cells after cerebral ischemical reperfusion injury: Brain tissues of rats oin each group be HE dyeing after 24 h ischemia reperfusion injury,to observe pathomorphology changes and calculate density of abnormal nerve cells in cerebral cortex and hippocampal.4. Observe Atorvastatin calcium's effect to rats' NF-κB expression and neuron apoptosis in brain tissues in each group. Immunohistochemistry and TUNEL were used to detect NF-κB expression and neuron apoptosis.ResultsThe expression of NF-κB in the ischemic area up-regulated after cerebral ischemia (P<0.01),peaking at 24 h after ischemia reperfusion injury. Concurrently,the number of apoptotic cells increased obviously(P<0.01).Atorvastatin-calcium could decrease the expression of NF-κB (P<0.01) and reduce neuron apoptosis,neurological deficit score(P<0.01),minificate rats'cerebral infarction volume,etc.ConclusionsAtorvastatin-calcium can relieve ischemia reperfusion injury of brain tissue and improve nerve function lose. The mechanism may be related with the decrease of NF-κB expression,thus decrease brain cell's apoptosis.
