Effects Of Jiaomu Oil On Mucus Hypersecretion In The Airway Of Asthmatic Rats

Objective To observe the effects of Jiaomu Oil on airway inflammation,airway goblet cell (GC),lung tissue Epidermal Growth Factor Receptor(EGFR) and serum levels of IL-13 and IL-4 in asthmatic rats and to explore its mechanism in the treatment of Mucus hypersecretion in asthma.Method(1) Thirty-two Male SD rats(200±20g) were randomly divided into four groups:â‘ Normal control group(n=8);â‘¡Asthmatic model group(n=8);â‘¢Prednisone group (n=8);â‘£Jiaomu Oil group(n=8).(2) Except the Normal control group,rats were sensitized with 1ml mixture of ovalbumin and AL(OH)₃ at the first day and the 8^th day.(3) Sensitized rats were treated with drugs once daily for 5 days from the 13^th day.Normal control group and asthmatic model group received normal saline in 10ml/kg/d;Prednisone group received Prednisone in10mg/kg/d;Jiaomu Oil group received Jiaomu Oil in 751.9mg/kg/d.(4) Except the Normal control group,rats in the rest of groups were challenged with ovalbumin for 20 minutes after one hour of received drugs in the 15^th,16^h and 17^th and were killed after 48 hours of challenge.While Normal control group were injected and challenged only with normal saline.(5) Their abdominal vein blood were collected for serum to detect the levels of IL-13 and IL-4.(6) The right lung were cut to make sections,then the lung tissue sections were stained with hematoxylin and eosin(HE) for general morphology and alcian blue-periodic acid-schiff(AB-PAS) for identification of goblet cells,and EGFR proteins were located in bronchial-pulmonary tissues using immunohistochemical analysis.Results The levels of IL-13 and IL-4 and ratios of GC were significantly decreased in the Jiaomu Oil group and Prednisone group than in the asthmatic model group(P＜0.01～P＜0.05). Airway inflammation and goblet cell hyperplasia(GCH)/goblet cell metaplasia(GCM) were significantly alleviated and no mucus retention in the airway lumen were observed in Jiaomu Oil group and Prednisone group,and mildly GCH/M were restricted in the larger airways.But no significantly difference of the expression of EGFR proteins in the lung in the Jiaomu Oil group and Prednisone group(P＞0.05). Conclusion Jiaomu Oil can alleviate the airway inflammation and suppress GCH in asthmatic rats by decreasing the levels of IL-13 and IL-4.Jiaomu Oil is valuable in the prophylaxis and treatment of asthma.While Jiaomu Oil can't suppress the EGFR proteins located in bronchial-pulmonary tissues.
