Objective The research mainly clarify the efficacy of the BaoShen decoction in preventing and treating the hyperlipidemia-induced renal damage and its mechanism through observing the influence to the hyperlipemia big mouse serum lipid(TC,TG,HDL-C),the kidney function(Scr,BUN),the kidney organization pathology(kidney index,the HE dyeing) and the immunohistochemistry(FN,TGF-β1) of renal cortex expressession,and lay the foundation for clinical application in pharmacology of the BaoShen decoction.Methods①high fat diet(ordinary feed adds 3%cholesterol,1%Cholic acid sodium,7.5% egg yolk powder and 10%Lard)feeds the Wistar big mouse to manufacture the model of the lipid-induced renal injury.②stochastically divided the big mouse into 6 groups,respectively be the normal control group(A group),the model control group(B group),the Simvastatin treatment group(C group),BaoShen decoction low dose group(D group),the BaoShen decoction middle dose group(E group),BaoShen decoction high dose group(F group),8 in each group,each group 12 weeks of treatment.③After the end of the experiment,examine the Blood Lipid separately(TC,TG,HDL-C),the kidney function(Scr,BUN),the kidney organization pathology(kidney index,the HE dyeing),and the kidney cerebral cortex immunity group(FN,TGF-β1).Results①Compared to the normal control group,the serum TC and the TG of the Model control group obviously advances(P<0.01),And the serum HDL-C of the Model control obviously reduces(P<0.01);Compared to the model control group and the Simvastatin treatment group,the serum TG of the BaoShen decoction high dose group reduce obviously(P<0.01);Compared to the model control group,the serum TC of each treatment group obviously drops(P<0.01),And the high dose group is obviously lower than the dosage, the low dose group(P<0.05,P<0.01);Compared to the modelcontrol group,the serum HDL-C of the BaoShen decoction high dose group obviously elevates(P<0.01) and is obviously higher than the low dose group(P<0.05).②Compared to the normal control group,the Scr and BUN of the model control group have the trend of escalation,but there is non-significance difference(P>0.05);Compared to the model control group,the Scr and BUN of each treatment group hasn't significance difference(P>0.05),but each of them,especially in the BaoShen decoction high dose group,has the declining trend.③Compared to the normal control group,the kidney index of the model control group obviously elevates(P<0.01), intercapillary Cells proliferation,mesenrium ground substance multiplication,mesangial region dilation,glomcrulus focal Segmented sclerosis by chance;Compare to the model control group,the kidney index of the BaoShen decoction high dose group and the middle dose group obviously reduce(P<0.01,P<0.05),The intercapillary Cells proliferation,the mesenterium ground substance multiplication and mesangial region dilation of each treatment group has relieved by different degree,but the high dose of Baoshen decoction is more obvious.④Compared to the normal control group,the FN of the model control group obviously advances(P<0.01);Compared to the model control group,the FN of the BaoShen decoction high dose group and the middle dose group obviously drops(P<0.01);Compared to the model control group,the FN of the BaoShen decoction middle dose group and the low dose group have the declining trend,but non-significance difference(P>0.05).⑤Compared to the normal control group,the TGF-β1 of the model control group obviously advances (P<0.01);Compared to the model control group,the TGF-β1 of the BaoShen decoction high dose group and the middle dose group obviously drops(P<0.01,P<0.05);The TGF-β1 of the BaoShen decoction high dose group is obviously lower than the low dose group(P<0.05).Conclusions The BaoShen decoction has the latent prevention hyperlipidemia-induced renal damage,improve the kidney pathological change,reduce the kidney index,suppress the production of the Renal Cortex FN and the expression of TGF-β1 through reducing the blood lipid and improving the lipid metabolism,the function that we have mentioned exist dose-response relationship,the high dose group to be most remarkable.
