Objective:Methylenetetrahydrofolate reductase (MTHFR) is a methionine-folate metabolism, a key enzyme, It can make 5,10-methylenetetrahydrofolate reduced to 5-methylenetetrahydrofolate in the body, as the main active form of folic acid in vivo. MTHFR activity can guarantee the normal metabolism of folate, DNA synthesis and methylation of the normal. MTHFR catalytic determine cell folate is way to enter or participate in methylation of nucleotide synthesis, The MTHFR gene mutation will affect the MTHFR enzyme activity and DNA synthesis and methylation, which constitutes a disease risk factor. This study analyzed the folic acid, vitamin B12, methylenetetrahydrofolate reductase (MTHFR) genotype groups in Yunnan province of breast cancer, benign breast disease and normal population distribution of the population difference, Explored the folic acid, vitamin B12, methylenetetrahydrofolate reductase (MTHFR) genotype and the relationship between breast cancer susceptibility.Methods:1. According to the principle of informed consent, fasting blood 5ml. Take 4ml blood serum separation and radioimmunoassay analysis of serum samples of patients with breast cancer and control the conten t of folate and vitamin B12.2. Fasting blood taken in accordance with the lml root Beijing tian-gen produced blood DNA extraction kit to extract nuclear DNA.3 Use ordinary PCR-RFLP technique,64 cases of breast cancer patients in Yunnan province,59 cases of benign breast disease and 62 normal people MTHFR 677 site 1298 polymorphism screening,64 cases of breast cancer and MTHFR genotype and estrogen receptor ER and progesterone receptor PR compared the frequency distribution of the situation.4 Compared folic acid, vitamin B12 locus in the MTHFR 677 CC CT TT three subgroups and 1298 points AA AC CC between the three sub-groups with or without significant differences in concentration levels.Result:1 The results show that, MTHFR677 mutation homozygotes (TT) in the tested breast cancer population (n= 64) and normal control group (n= 62) and benign patient group (n= 59) in the distribution of frequencies were 9.38%,18.75% and 16.13%, no significant difference; MTHFR 1298 mutation homozygotes (CC) subjects in the corresponding breast cancer population (n= 64) and normal control group (n= 62) and benign patient group (n= 59) in the distribution of frequencies were 3.13%, 10.17% and 3.23%, no significant difference. In the current sample conditions, MTHFR 677 and 1298 genotype polymorphism and breast cancer population in Yunnan province was no significant correlation between the susceptibility.2 In 64 cases of breast cancer case group and 677 in the estrogen receptor genotype x2 = 74.86> x~20.005,2, p<O.005, shows a relationship between the two in the estrogen receptor and the 1298 genotype between x~2 = 69.11> x~20.005,2, p<0.005, shows a relationship between the two, PR and 677 in genotype x~2= 69.34> x~20.005,2, p 0.005, shows a relationship between the two, in the progesterone receptor and 1298 genotypes x~2= 74.70> x~20.005,2, p<0.005, shows a relationship between the two. Two patients of breast cancer Vitamin B121298CC group was significantly lower than among other people groups. Between groups in other folate and vitamin B12 levels were not significant differences. Conclusion:This study has not yet proved that MTHFR gene single nucleotide polymorphisms associated with breast cancer have an increased risk, but with low folate intake and (or) metabolic disorder folic acid may play a role in breast tissue, especially in breast cancer 677TT group and 1298CC group, folic acid and vitamin B12 are particularly important. To further explore the pathogenesis of breast cancer provides a new direction for breast cancer prevention offer new ideas, because individuals carrying MTHFR gene mutation, more folic acid to maintain normal cell function, for these people to add folic acid possible role in the prevention of breast cancer. MTHFR genotype and estrogen receptor ER and progesterone receptor PR case the relevance of the frequency distribution of the research for the better correlation between folate and breast cancer provide a reference.
