Objective Nasopharyngeal carcinoma (NPC) is one of Head and Neck malignancies and occurs at a higher incidence in Southwest China.NPC usually connects with changes in cell cycle and apoptosis mechanisms caused by some gene mutation. Our object is to evaluate the application of RNA interference in the study of survivin gene in nasopharyngeal carcinoma(NPC)cell line CNE2.Observe the effects of survivin silencing on NPC cell growth.Methods Designed the specific small interference RNA(siRNA)of the gene sequence of survivin and transfected into NPC cell using OligofectamineTM reagent. mRNA and protein levels of survivin were detected by semi-quantitative reverse transcriptase polymerase chain reaction (RT-PCR)and Western blot. Flow cytometry and MTT assay were employed to analyze the effects on cell cycle and proliferation.Results The specific siRNA of the gene sequence of survivin can suppress the survivin expression efficiently and stably in CNE2 and down regulation of survivin resulted in significantly inhibition of tumor cell growth in vitro. At 24 h and 48 h,after the transfection, the growth inhibitory rates were significantly higher in specific Survivin siRNA group than those in the control. While G0/G1 phase arrest was shown.Conclusions These results provided solid testimony that survivin gene was vulnerable to RNA interference and, selective inhibition of survivin had anti-proliferation effect on NPC cell. Inhibition of the expression of survivin can induce apoptosis of NPC cell.RNA interference could be a powerful tool in further investigations of survivin and a novel therapeutic strategy for NPC patients.
