[Objective]:To study the express of p27kip1 and JAB1 in carcinoma of colon and their mutual relationship,then to investigate their roles and probable mechanism in the pathogenesis of colon cancer.[Methods]:Immunohistochemistry was used to detect the expression and distribution of p27kip1 and JAB1 in carcinoma of colon and normal colon.Statistics was used to analyze the data.[Results]:The expression of protein p27kip1 and JAB1 protein was mainly localized in the cytoplasm,nucleus as well as some expression.The positive expression rate of p27kip1 protein in colon cancer was significantly lower than that in adjacent normal colon(χ~2=5.214,v=1,p<0.05).The expression of p27kip1 protein was different in different differentiation grades of colon cancer(χ~2=7.127,v=2,p<0.05).The expression of p27kip1 in lymph node metastasis of colon cancer was higher than that in the absence of lymph node metastasis group(z=2.849615,p<0.05).The expression of p27kip1 inⅠ-Ⅱstages was higher than that inⅢ-Ⅳstages(χ~2=6.353,v=1,p<0.05). The expression of p27kip1 protein was no significant difference betweenⅠandⅡstages(p>0.05).The expression of p27kip1 protein was no significant difference betweenⅢandⅣstages(p>0.05).The positive expression rate of JAB1 protein in colon cancer was significantly higher than that in adjacent normal colon(χ~2=13.181,v=1,p<0.05).The expression of JAB1 protein was different in different differentiation grades of colon cancer(χ~2=11.07,v=2,p<0.001).The expression of JAB1 in lymph node metastasis of colon cancer is higher than that in the absence of lymph node metastasis group(z=2.157268,p<0.05).The expression of JAB1 inⅠ-Ⅱstages was lower than that inⅢ-Ⅳstages(z=-2.157268,p<0.05).The expression of JAB1 protein was no significant difference betweenⅠandⅡstages (p>0.05).The expression of JAB1 protein was no significant difference betweenⅢandⅣstages(p>0.05).JAB1 proteins and p27kip1 protein had the negative correlation in colon cancer(r_s=-0.5885,p<0.001).[Conclusion]:1.The expression of p27kip1 protein had a significant difference between colon cancer and normal colon tissue.The positive expression rate of p27kip1 protein in colon cancer was significantly lower than that in adjacent normal colon,p27kip1 protein as a tumor suppressor factor that can regulate cell proliferation,differentiation and apoptosis of ability.Low expression of p27kip1 protein led to tumor occurrence and development in organization.2.The expression of JAB1 protein had a significant difference between colon cancer and normal colon tissue.The positive expression rate of JAB1 protein in colon cancer was significantly higher than that in adjacent normal colon.JAB1 protein play an important role in the development of colon cancer.3.There was a negative correlation between JAB1 protein which in colon cancer with high expression and p27kip1 protein with low expression.There was a correlation between JAB1 protein,p27kip1 protein and differentiation of colon cancer cell invasion and metastasis.JAB1 protein had been involved in colon cancer cell proliferation,differentiation,invasion and metastasis by making negative regulation with the cell cycle inhibitor of p27kip1 protein.4.Protein of p27kip1 and JAB1 would be considered as new clinical index of colon carcinoma,and make for diagnose of neoplasm differentiation,neoplasm metastasis and TNM staging.5.Depth JAB1 protein pathway involved in the mediation p27kip1 protein factor in the development of colon cancer in the role that will help to further understand the mechanism of colon cancer and provide some experimental evidence in colon cancer treatment.6.JAB1 protein as a new target for cancer gene therapy,the specific regulatory mechanism is unclear,pending further exploration.
