ABSTRACTRadix asparagi is the root of Asparagus cochinchinensis(Lou.)Merr.which is a kind of liliaceae perennial climber herbs.As one of traditional Chinese medicines, Radix asparagi has a lot of activity,such as anti-aging,anti-bacterial,anti-inflammatory, hypoglycemic and anti-tumor.The main contents in Radix asparagi include asparagines,phlegm,β-sterols,5-methoxy-methyl furfural and kinds of amino acids,and polysaccharide is also a very considerable proportion.There are many rearch for the Asparagus neural polysaccharides,but Aspartic acid polysaccharides rarely been reported.In order to fully develop the use of Asparagus polysaccharide,A-spartic acid polysaccharide was studied about extraction,separation,purification,and the immune and antioxidant activity in vitro of Aspartic acid polysaccharide were studied preliminarily.These results have provided a foundation of further research for Aspartic acid polysaccharide.The main results are as follows:1.In order to optimize the best conditions for the extraction process of polysaccharide from Radix asparagi,the celluase was used in the experiment.The effect of enzyme concentration,extraction time,temperature,pH and solid to liquid ratio on the pick-up rate of polysaccharide was considered.By L53 orthogonal test, the optimal conditions for polysaccharides extraction process are in the case of 0.8%enzyme,time about 90min,the temperature at 50℃,pH 5.2 and dosage liquor ratio 1:25.Under these conditions,the pick-up rate of polysaccharides was 6.922%.2.on the basis of the optimal extraction process,the active carbon was used to decolorize for the polysaccharides,and the optimum cond.itions include 1.8%carbon, pH 8.0,the temperature 40℃,time 1h.The result has showed that the decolorization 3 rate was 38.16%.3.Infrared spectroscopy (IR) suggested that the aspartic acid polysaccharides was a kind of pyranose with D(+)β-configuration.4.The Nick coral pink chicks were chosed to feed as investigatory animals and the Newcastle disease vaccine was selected to use as targeted vaccine.Under these conditongs,we have studied the immune adjuvant effects of the aspartic acid polysaccharides.The result showed that the polysaccharides could promote the secretion of the antibody, T cell number and immune organ index. So the aspartic acid polysaccharides have a certain effect of immunological enhancement.5.The chemical model systems were used to study the antioxygenation of the aspartic acid polysaccharides,in vitro.The study found that it had a certain reducibility and ability to remove hydroxyl radical and superoxide anion.
