In the study, the technology, extraction total flavones from Herba Hedyotids Chrysotridhae, was optimized. The condition was determined, which is technology of purification total flavones with macroporous resin and polyamide. Trichoderma, a kind of fungi, was chose to ferment Herba Hedyotids Chrysotridhae and the effect of ferment on extraction process total flavones was studied.According result of single factor experiment, fixing on appropriate levels, the result of the effects of times, ethanol concentration, extracting time, radio of liquid to material on extracting was studied. By mean of variance analysis, in these factors, ethanol concentration is the most significant factor, next came extraction times and extracting time then. The optimum extraction technology was as follow: concentration of Ethanol at 70%, ratio of liquid to solid at 16:1, extraction for 2 times and 1.5 hours each time. The repeatability test shows that this technology has good repeatability and is reliable. In the extraction of this technology, the mass fraction of flavones is 12.32%.The NKA-9 type resin is chose from six macroporous resins to Preli minary purify total flavones compounds. Preli minary purification process, determined by the dynamics adsorption test, was follow that at the flow rate of 1.5 mL/ min, the extraction solution of concentration is 2.5 mg/mL was loaded onto the column, then 10BV of water was used to elute at flow rate of 1.5 mL/ min to remove polysaccharide and its pH is 5, and the last 3BV of 70% ethanol was used to elute at flow rate of 2.0 mL/min. By Preli minary purification, the flavones mass fraction was raised 20.33% to 32.65%, but its purity is not high and need further purification.Advantages and disadvantages between the polyamide resin and silica gel were compared by the static adsorption test and found that the polyamide resin is more suitable to purify flavones from Herba Hedyotids Chrysotridhae. The dynamics adsorption test showed that 10BV water of its pH=5 was used to remove polysaccharide and 4bv ethanol of the concentration of 50% was used to eluted. Purification by mesas of the polyamide resin has achieved a good effects and the flavones mass fraction was raised from 16.0% to 48.53%.In the study, Trichoderma was chose from 5 kind fungi to ferment Herba Hedyotids Chrysotridhae. After fermentation, the extract rate and yield rate of flavones was obviously raised, which show that Trichoderma fermentation have promotion function on extracting total flavones from Herba Hedyotids Chrysotridhae.
