Drug Releasing Test In Vivo And In Vitro Of Alginate/Chitosan Incorporated With Vancomycin As Drug Delivery System

bjective:To investigate the releasing profile and different factors effected in vitro in ALM/VCM releasing,and to compare and evaluate the difference between the methods of drug delivery system and systematic injection.Methods:Four types of VCM/ ACA were first prepared:different ratio of powder to mixing solution(5:6,1:1) under freezing and drying or not.Then each group of VCM/ACA was putting in bottle dipping and to observe the releasing characteristic.The experimental middle of left femur bone defect models in rabbits were first prepared.Then the animals were divided into two groups, In group A with VCM/ACA inlaid to the defects the drug concentrations of the serum,the local bone and the muscle at 0.5h,1h,6h,24h,72h,1w and2w after operation were determined. In group B with VCM solution injected into the rabbit ear vein,the drug concentrations of the serum,the local bone and the muscle at 10min,0.5h,1h,6h,24h,72h,1w and 2w after injection were determined too.In addition,histologic sections of body tissues were made to look for signs of systemic toxicity of the implant.Results:VCM/ACA can lasting more than 250 hours.The releasing characteristic is dependent on drug amount,mixing solution volme and under freezing and drying.Drug delivery system could provide a high local concentration level which sustained for at least two weeks,while VCM concentration in serum was much lower at the same time.And histologic analysis of multiple tissues revealed no histologic evidence of toxicity.Conclusion:The VCM/ACA(1:1) under freezing and drying has remarkable relseasing profile,has feasibility and safety in vivo in course of ALM/VCM releasing.That is what we need in the treatment of clinical,can use to satisfy the need of clinical.
