Objective By observing the experimental mice oral ulcer and peripheral blood serum IL-2, IFN-Γlevel, to evaluate Buzhongyiqitang therapeutic effect in the spleen deficiency type recurrent oral ulcers mice, discuss the effect mechanism, in order to provide a theoretical basis for clinical application.Method (1) Use restricted diet and excessive fatigue way to establish a mice model of spleen deficiency, on this basis, inject with antimetabolite chemotherapy drug- intraperitoneal methotrexate, once a day, continuous for five days. Joint use the three methods to reproduce the model of spleen deficiency type recurrent oral ulcers mice successfully. (2) Detect normal mice and model mice peripheral blood of IL-2, IFN-Γlevel. Check normal and damaged oral mucosa, and do pathological examination. (3) Then divide remaining model mice into Buzhongyiqitang treatment group and saline treatment comparison group randomly, both groups give normal diet, observing general symptoms and the oral ulcers change of mice in each group. After 10 days treatment, test IL-2, IFN-Γcontent.Results Observation of oral mucosa pathological organization showed that: the normal group mice's oral mucosa is smooth, no significant inflammatory cell infiltration. Model group mice's Oral mucosal ulcers can see- there is necrosis of mucosa epithelium, detached, surface coverage of a large number of neutrophils and fibrous exudation, mucous membrane with infiltration of inflammatory cells. Model mice serum IL-2, IFN-Γlevels decreased significantly (P <0.01) than normal mice. Compared Buzhongyiqitang treatment group with saline treatment comparison group, two groups of mice peripheral blood saline level of IL-2, IFN-Γhave increased, the treatment group compared with the normal group is not much difference (P> 0.05), but saline treatment comparison group compared with the normal group have differences (P <0.01). Treatment group IL-2, IFN-Γlevel increased quickly than the comparison group , the two groups were significantly different (P <0.01).The comparison group compared with the model group is not much difference (P> 0.05).Conclusion 1 Buzhongyiqitang can improve spleen deficiency symptoms significantly, promote ulcer healing and reduce symptoms, reduce ulcer recurrence.2 Buzhongyiqitang can increase IL-2, IFN-Γ, enhance immune function.3 The occurrence of spleen deficiency type recurrent oral ulcer has important relationship with decreased immune function.
