The Preliminary Study On Killing Osteosarcoma Cell Line(MG-63) By ¹²⁵I Seed

Objective:Study on killing osteosarcoma cell line(MG-63) by ¹²⁵I seed,and understand the killing effect which in logarithmic growth phase,provide theoretical basis for endovascular radiation trentment of osteosarcoma postoperative.Further improve the postoperative limb salvage rate and 5- year survival rate,so as to reduce the recurrence and metastasis rate.Methods:Choose the osteosarcoma cell to culture in culture dish with 35 mm in diameter,when the cell count＞10⁶/mL.The number of ¹²⁵I seed placed at the botton of each Culture dish was 8,one was placed in middle,and the control group was not irradiated.All the culture dishes were placed in 37℃thermostat.The specimens were obtained on day 1,2,3 after irradiation.Analysis of apoptosis rate of osteosarcoma cell line with flow cytometry(24h,8h,72h).Electron microscopy analysis of cell morphology.The tested data are performing randomized block analysis of variance.Results:Osteosarcoma cell line were exposed by continuous low-dose irradiation of ¹²⁵I seed.Analysis of apoptosis rate of osteosarcoma cell line with flow cytometry. The results showed that the apoptosis rate of the control group(24h,48h,72h,n=6) was(0.87±0.09,0.90±0.17,1.04±0.22),and the experiment group was(4.98±0.53,6.76±0.39,10.50±0.56).The tested data are performing randomized block analysis of variance showed that tumor cells in the control group were arranged densely,cell morphology was anomalistic.The nucleo cytoplasmic ratio was increased. There can be several nucleolus,other organelles have no tumefaction;each stage of apoptosis can be seen in the experiment group.such as residual body,cell nuclear chromatin edge gathered,cell nuclear chromatin areintensely concentrated,apoptotic bodies.Conclusion:1,Killing osteosarcoma cell line can be Killed by ¹²⁵I seed.2,The killing effect increased with the radiation time prolonged.
