| ã€Objective】To study the effect of endostatin on growth inhibition, induction of apoptosis and cell cycle of nasopharyngeal carcinoma cell CNE2 while combined with radiotherapy and to find out experimental feasibility of endostatin radiosensitivity.ã€Method】The inhibition effect of different concentrations of endostatin on nasopharyngeal carcinoma cell CNE2 was observed with colorimetric test (MTT) and the optimal concentration was calculated. Time inhibition of endostatin combined with radiotherapy on nasopharyngeal carcinoma cell CNE2 was detected with MTT and the effection to cell apoptosis and cell cycle detected with flow cytometry.ã€Results】1. Endostatin treatment, degree of inverted microscope and nasopharyngeal carcinoma cells after treatment was found with the drugs and the role of time increased, CNE2 cells became round and floating, refraction is low and loosely connected cells, proliferation slow down.2. MTT assay different concentrations of ex degrees at different time points on CNE2 growth inhibition effect of different concentrations in the nasopharyngeal carcinoma cell Endostatin CNE2, with the inhibitors concentration, treatment time, tumor cell viability gradually lower.3 Combined therapy showed the strongest effect to nasopharyngeal carcinoma cell CNE2 single radiation dose 4Gy and 100mg/l concentrations of endostatin. Combined therapy was more effective than that single endostatin or single radiation. Tumor cell survive rate decreased with time passed.4. Flow cytometry analysis showed that:human nasopharyngeal carcinoma cells apoptosis with fine CNE2 G2/M phase percentage was negatively correlated with degree of grace may increase the cell G2/M radiosensitivity. 5. TdT-mediated dUTP nick-end labeling (TUNEL), detecting apoptosis:Endostatin concentration 100mg/l, single dose 4gy, function 72h, to PI double staining flow cytometry, and the apoptosis rate necrosis rate in the control group was only 1.7% apoptosis, cell necrosis was 8.1% in the experimental group, the apoptotic rate was 11.6%.ã€Conclusion】1. The determined concentration of endostatin had inhibition effect on human nasopharyngeal carcinoma cell line CNE2 and it was time and concentrations dependence.2. The radial sensitizing effect of endostatin combined with radiotherapy appeared time dependence. |
