Objective:To study the relationship between the relative quantity of FSHR and SCF expression and the spermiogenesis,the quantity expression of testicular tissue of patients with obstructive and non-obstructive azoospermia and its differences were analyzed preliminarily.Methods:36 testicular paraffin samples,from 2007 to 2009,with azoospermia were divided into two groups:obstructive and non-obstructive azoospermia,according to the pathological results.The quantity of expression of FSHR and SCF were tested by immunohistochemistry and integral optical density of the computer graphics analysis system.19 cases with azoospermia from August 2008 to April 2010 were divided into two groups:obstructive and non-obstructive azoospermia according to the pathological results.11 samples of which was examined with immunohistochemistry. The quantity expression of FSHR mRNA and SCF mRNA were tested with the real time PCR.The original relative expression quantity of mRNA and differences in the two groups were indicated with 2-△CT.Results:①Immunohistochemistry:There were 21 cases in the group of the obstructive azoospermia and 15 cases in the the group of the non-obstructive azoospermia.The mean and standerd error of the integrated optical density values of the FSHR in obstructive azoospermia group and non-obstructive azoospermia group were 49.525±16.626 and 33.3927±9.916,respectively.(P=0.002).The mean and standerd error of the integrated optical density value of the SCF in obstructive azoospermia group and non-obstructive azoospermia group were 13.717±5.664 and 10.2747±3.137,respectively.(P=0.026).The quantity of expression of FSHR and SCF in the obstructive azoospermia group were significantly higher than that of the non-obstructive azoospermia group.There was statistically significant difference between the obstructive and non-obstructive azoospermia group(P<0.05).②Real time PCR:There were 8 cases in the group of the obstructive azoospermia and 11 cases in the group of the non-obstructive azoospermia.The mean and standerd error values of the 2-△CT of the FSHR mRNA in obstructive azoospermia group and non-obstructive azoospermia group were 0.0015±0.00101 and 0.0006±0.00059,respectively.(P=0.022).The mean and standerd error values of 2-△CT of SCF mRNA in obstructive azoospermia group and non-obstructive azoospermia group were 0.0028±0.00215 and 0.0009±0.00075,respectively.(P=0.012).The quantity expression of FSHR mRNA and SCF mRNA in the obstructive azoospermia group was higher than that of the non-obstructive azoospermia group.There was statistically significant difference between the obstructive azoospermia group and non-obstructive azoospermia group(P<0.05).Conlusion:The down-regulated quantity expression of FSHR and SCF mRNA in the testical of non-obstructive azoospermia might one of the causes of differentiation dysfunction in the azoospermia patients.
