Objective Nerve stem cells from the whole brain of newborn Sprague Dawley rats were differentiated into dopaminergic neurons in vitro.Then the dopaminergic neurons were transplantated into rat model of Parkinson disease.we studied in vivo the survival and biological function of those cells and evaluated therapeutical effects for rat.Methodsâ‘ Rat models of PD were established by injecting 6-OHDA into the right substantia nigra of rats midbtain.â‘¡Neural stem cells were isolated and cultured,then were induced by 400nmol/lAII for 14 days.The expression of TH were detected by immunocytochemical technique.â‘¢The differentiated cells were transplanted into striatum of the eligible rats. The rotary behavior induced by APO was observed at 2,4,6 weeks.In the striatum of rats, the expression of TH,DAT and distribution of positive cells were detected by immunocytochemical technique .Resultsâ‘ Among experimental 60 rats which were injected APO, 32 rats showed steady rotation to uninjured side(ï¹¥7r/min) at 36 days after the right substantia nigra of rats were damaged by 6-OHDA.â‘¡Induced by 400nmol/L AII, neural stem cells can be differentiated into dopaminergic neuron.â‘¢The rotational asymmetry was decreased at 42 days after the differetiated cells were transplanted into PD rats( P < 0.05). Immunohistochemistry showed that there were TH,DAT optisive neural cells and those cells mainly located in borderline of pinhole in the striatum of rat.Conclusion Neural stem cells were differentiated dopaminergic neuron and were transplantated into striatum of PD rats.Those cells can survive in the striatum and improve the motor function of PD tats.
