OBJECTIVE:To study mechanism of Endostatin influenced on hepatoce-llular carcinoma by its inhibiting proliferation of SMMC-7721 human heaptoce-llular carcinoma cells and nude mice planting hepato-cancer and expression of their VEGF,VEGFR,uPA,uPAR and MMP-2.METHODS:SMMC-7721 human hepatocellular carcinoma cells were cu-ltured.Inhibited effect of Endostatin on the proliferation of SMMC-7721 human hepatocellular carcinoma cells was detected by MTT.An animal model was esta-blished through inoculating subcutaneously human SMMC-7721 hepatocellular carcinoma cells into nude mice. Inhibited effect of tumor growth of nude mice bearing cancer in different densities of Endostatin was detected. Expression of VEGFmRNA,VEGF-CmRNA,VEGFR-1mRNA, VEGFR-2mRNA, uPAmRNA and uPARmRNA in human SMMC-7721 hepatocellular carcinoma cells and tumor tissue of nude mice planting cancer was detected by RT-PCR. Protein expression of VEGFR-1,VEGFR-2 and MMP-2 in tumor tissue of nude mice planting cancer was detected by western blot.RESULTS:In vivo,weight and volume of nude mice planting hepato-can-cer in the Endostatin groups was smaller than in the control group(P<0.05).Ex-pression of VEGFmRNA,VEGF-CmRNA,VEGFR-1mRNA,VEGFR-2mRNA, uPAmRNA and uPARmRNA in Endostatin groups was lower than control group (P<0.05).Protein expressions of VEGFR-1, VEGFR-2, MMP-2 in the Endostati-n groups was lower than in the control group(P<0.05).In vitro, there is no signi-fycant difference of inhibition ratio and expression of VEGFmRNA in human SMMC-7721 hepatocellular carcinoma cells between Endostatin groups and co-ntrol group(P>0.05).CONCLUSIONS :Endostatin has a sigificant depressant effect on growth of mude mice planting hepatocellular carcinoma. Endostatin may obviously in-hibit growth of hepatocellular carcinoma by blocking the signal conductive pa-thway of VEGF/VEGFR. Endostatin may inhibit metastasis and infiltration of hepatocellular carcinoma by inhibiting expression of uPA,uPAR and MMP-2.
