ObjectivesTo investigate the expression of Smad2/3,Smad4 and Smad7 in human endometrial carcinoma and their clinical-pathological significance.MethodsSmad2/3,Smad4 and Smad7 were measured in normal endometriuim,atypical hyperplasia endometrium and endometrial carcinoma by SP immunohistochemical method.Results1. The positive rate of expression of Smad2/3,Smad4 and Smad7 in human endometrial carcinoma was significantly lower than normal endometrial tissue and atypical hyperplasia of endometrial tissue, from normal endometrial tissue to atypical hyperplasia endometrium, to endometrial carcinoma, it was a steady decline,the difference was statistically significant (P<0.05)2. In endometrial carcinoma, the expression of Smad2/3,Smad4 and Smad7 was not related to surgical-pathological staging (P>0.05); the expression of Smad2/3 and Smad7 was related to histological grading:with the increase of histological grade the expression decreased (P<0.05)3. There was a positive correlation between Smad2/3 and Smad4 in endometrial carcinoma (P<0.05); There was no relation between Smad2/3 and Smad7 in endometrial carcinoma (P>0.05) Conclusions1.Smad2/3,Smad4 and Smad7 were all involved in the process of endometrial carcinoma.2. In endometrial carcinoma,the loss expression of both Smad2/3 and Smad7 had some connection with the differentiation of endometrial carcinoma.Tthe expression of Smad4 was not directly related to the degree of malignancy of endometrial carcinoma, it may not be involved in the invasion and evolution of endometrial carcinoma when it occurred.3. There was a positive correlation between Smad2/3 and Smad4 in endometrial carcinoma suggests that they may co participate in the pathogenesis and development of endometrial carcinoma through interacting with each other, detecting the expression of Smad2/3 and Smad4 in endometrial carcinoma has an important value in evaluating the degree of malignancy and prognosis of endometrial carcinoma. There was no relation between Smad2/3 and Smad7 in endometrial carcinoma,the interaction is uncertain in pathogenesis and progression of endometrial carcinoma.
